The Effect Of Medicinal Scraping On The Expression Of Caspase-3, Caspase-9 And XIAP In Chondrocytes Of Rabbit KOA Model

Objective: By observing the expression of caspase-3,caspase-9 and XIAP in rabbit chondrocytes of KOA model,the effects of tiebaojin massage cream combined with scraping method on the apoptosis of rabbit chondrocytes of KOA model were investigated from the mitochondrial/cytochrome c-mediated apoptosis pathway,so as to further verify the clinical efficacy of scraping method in the treatment of KOA and explain part of its mechanism.Methods: Forty 6 month old New Zealand rabbits were randomly divided into the normal group,the model control group,the tiebaojin massage cream combined with scraping method group,the tiebaojin massage cream group and the scraping group,with 8 rabbits in each group.In animal experiments,papain articular injection was used to model the osteoarthritis of knee in rabbits.After the intervention of rabbit K0 A model in each group,the expression of caspase-3,caspase-9 and XIAP proteins in the articular cartilage of knee osteoarthritis in rabbits was detected by immunohistochemistry.Results: After intervention,the cartilage histomorphology observation results of each group were scored.Compared with the normal group,the scores of tiebaojin massage cream combined with scraping method group,tiebaojin massage cream group,scraping group and model group were significantly higher,with significant differences(P<0.05).Compared with the model group,the Mankin scores of the tiebaojin massage cream combined with scraping method group,tiebaojin massage cream group and the scraping group were lower,and the difference was statistically significant(P<0.05).Compared with the tiebaojin massage cream combined with scraping method group,there was significant difference between the tiebaojin massage cream group and the scraping group(P<0.05).It is suggested that the tiebaojin massage cream combined with scraping method,tiebaojin massage cream,scraping can effectively delay the cartilage degeneration of rabbit model of knee osteoarthritis,and the effect of traditional Chinese medicine scraping is more obvious.The expressions of caspase-3,caspase-9 and XIAP on chondrocytes were detected by immunohistochemical assay after intervention,and the results were as follows: caspase-3 and Caspase-9: compared with the normal group,the expressions of the scraping group,the tiebaojin massage cream group,the scraping group and the model group were significantly increased,with statistically significant differences(P<0.05).Compared with the model group,the expressions of scraping group,tiebaojin massage cream group and scraping group reduced(P<0.05).Compared with the medicine scraping group,the expression of tiebaojin massage cream group and scraping group increased more(P<0.05).XIAP: compared with the normal group,the expressions in the scraping group,the tiebaojin massage cream group,the scraping group and the model group were significantly reduced,with statistically significant differences(P<0.05).Compared with the model group,the expressions of scrapinggroup,tiebaojin massage cream group and scraping group increased(P<0.05).Compared with the scraping group,the expression of tiebaojin massage cream group and scraping group decreased(P<0.05).It is suggested that the use of drug scraping,tiebaojin massage cream and scraping can significantly reduce the expression of caspase-3 and caspase-9 in cartilage tissue,increase the expression of XIAP,reduce the damage to cartilage,protect cartilage tissue,and delay the degeneration of KOA joint.Its traditional Chinese medicine scraping method is more effective.Conclusion:1.scraping method,tiebaojin massage cream and the tiebaojin massage cream combined with scraping method can effectively improve the cartilage degeneration of rabbit knee osteoarthritis model,but the last one works better.2.The tiebaojin massage cream combined with scraping method can reduce the expression levels of caspase-3 and caspase-9 in the articular cartilage of K0 A model rabbits,increase the content of XIAP,thus protecting the articular cartilage from damage and delaying joint aging.This may be one of the mechanisms by which the tiebaojin massage creams are combined with scraping to treat KOA.
