Objective:This study was designed to discuss the machanism of MMP-9/NOS-2 in experimental allergic neuritis,and evaluate the effect of extracorporeal whole blood immunoadsorption on it.Methods:EAN Wister rats were set up by immunization of MBP.MBP was extracted from bovine sciatic nerve.The EAN rats were divided into three groups based on the development of the symptoms,e.g.experimental group of symptom beginning/climax/recovery.The clinical features were quantitative according the Gabyriel clinical scale system.HE and Masson stain else the immunochemistry of MMP-9/NOS-2 were done on the sciatic nerve section.Electrophysiological characteristics of EAN rats on day 15 post immunization and normal rats were assessed by examining the conduction velocity and latency in sciatic nerves.On the day 13 post immunization,EAN rats were treated by NS or extracorporeal whole blood immunoadsorption.And a series of assessment,e.g.the clinical scale, pathology,EMG,MMP-9 were done on day 18 post immunization.Results:The extracting of the sciatic nerve were analysed by SDS-PAGE.Three bands were found,which matched with the P0,P1,P2 protein.The concentration of extracting protein detected by Folin was 7 or 20mg/ml respectively.And around 100mg of MBP could be achieved from 100g bovine sciatic nerve.The symptoms of the EAN rats showed on day 7-9 post immunization,developed to the climax on day 13-18p.i.,with the clinical score 10-12,recovered on day 22-25 p.i;and no abnormality found in the control group.The T cell number of three experimental groups and control group was 8.54±1.06,12.8±1.04,5.12±1.45,0.0±0.00.Masson stained showed demyelination of EAN rats,and correspondence with the symptoms. The nerve conduction velocity and latency of EAN group obviously decreased.The cells expressed MMP-9 were detected on the blood vessel and Schwann cell,and the comparison of the EAN group at the symptoms onset and climax with the control group,there were significant differences.The T cell expressed NOS-2.After undertaking the extracorporeal whole blood immunoadsorption,the weight of the EAN rats showed recovering on day 18 after immunization,and almost became normality on day 22-24,and at the meantime,the number of T cells,the demyelination and the EMG became better,compared with the EAN without treatment,p<0.05.Conclusion:MBP can successfully set up EAN rats.During the development of the EAN,MMP-9 involved in the damage of the blood-nerve barrier and the degradation of myelin.And extracorporeal whole blood immunoadsorption had therapeutical effect on EAN rats.
