PARTâ… THE LEVELS OF HIGH SENSITIVE C-REACTIVE PROTEIN IN TYPE 2 DIABETIC FAMILY MEMBERSâ… a THE VARIATION AND INFLUENTIAL FACTOR OF HIGH SENSITIVE C-REACTIVE PROTEIN IN TYPE 2 DIABETIC FAMILY MEMBERSObjective To explore the variation and influential factor of high sensitive C-reactive protein in type 2 diabetic family members. Methods (1) We studied 939 subjects(426 male and 513 female) from Chongqing Han families. 804 of them came from T2DM family, and 135 of them were their spouses. Anthropometry parameter, blood pressure, renal function parameter, oral glucose tolerance test(OGTT), lipid levels, high sensitive C-reactive protein(hsCRP) were examined. T2DM were defined according to WHO diagnostic criteria. (2) All the subjects were divided into three groups: T2DM, first degree relatives(FDR) and normal control group. The clinical and biochemical indicators were compared among three groups. The SPSS statistics software was applied to analyse the dependability of the indicators.Results After adjusted for age and BMI, the levels of hs-CRP in T2DM and FDR group were higher than those in controls(P<0.05), and hsCRP level in T2DM group was higher than that in FDR group. Contrast to normal controls, the levels of BMI, SBP, WHR, AER, GLU0h, GLU2h, INS0h, Homa-IR and TG in T2DM and FDR group increased significantly(P<0.05), and HDL decreased significantly(P<0.01). Although there were higher thigh circumferences and INS2h levels in T2DM and FDR group than in control group, there was no statistics difference among three groups. The serum hs-CRP levels in T2DM and FDR groups were positively associated with the levels of BMI, WHR, abdominal circumferences, GLU2h, INS0h, INS2h, Homa-IR, TG and Cre, and inverse correlation with HDL levels. In FDR group, SBP and Homa-B levels were positively associated with the serum hs-CRP level.Conclusions The non-diabetic FDR of T2DM existed inflammatory reactions similar to T2DM. Accompanied to the aggravation of glycometabolic disturbance, inflammatory reaction aggravated gradually. It indicated that inflammatory response might play an important role in the pathogenesis of T2DM. â… b THE ASSOCIATION BETWEEN HIGH SENSITIVE C-REACTIVE PROTEIN AND INSULIN RESISTANCE IN IMPAIRED GLUCOSE REGULATION FIRST-DEGREE RELATIVES OF TYPE 2 DIABETES.Objective To explore the association between high-sensitive C-reactive protein and insulin resistance in impaired glucose regulation first-degree relatives(FDR) of type 2 diabetes.Methods (1) We studied 287 subjects from Chongqing Han families which were enrolled . 157 of them were FDR of T2DM , and 130 of them were the spouses of T2DM family members. Anthropometry parameter, blood pressure, renal function parameter, oral glucose tolerance test(OGTT), lipid levels, high sensitive C-reactive protein(hsCRP) were examined. IGR were defined according to World Health Organization (WHO) diagnostic criteria. (2)All the subjects were divided into four groups: impaired fasting glucose(IFG) FDR, impaired glucose tolerance(IGT) FDR, IFG+IGT FDR and normal control group. The clinical and biochemical indicators were compared among the four groups. The SPSS statistics software was applied to analyse the relations between hs-CRP and IR and the dependability of the indicators.Results After adjusted for age , the level of hs-CRP in IGT FDR or IFG+IGT FDR group was higher than that in controls(P<0.05), and hsCRP level in IFG+IGT FDR group was higher than that in IFG FDR or IGT FDR group. Contrast to IFG FDR, the level of hs-CRP in IGT FDR didn't increase significantly(P>0.05). The serum hs-CRP level in IGR first-degree relatives was positively associated with age, BMI, blood pressure, WHR, abdominal circumferences, OGTT 2h glucose, fasting insulin, OGTT 2h insulin, Homa-IR, TG, LDL and Homa-B, and inversely correlated with HDL.Conclusions The IGR first-degree relatives of T2DM existed conspicuous inflammatory reactions similar to T2DM. Inflammatory reactions correlated closely with insulin resistance. Inflammatory reactions and insulin resistance were more severe in IFG+IGT FDR. It indicated that dangerous heart-blood incidents occurred more frequently in IFG+IGT FDR. PARTâ…¡THE ASSOCIATION OF ADIPONECTIN GENE POLYMORPHISM WITH HIGH SENSITIVE C-REACTIVE PROTEIN IN TYPE 2 DIABETES FAMILIESObjective To investigate the relationship of adiponectin gene polymorphism and the high sensitive C-reactive protein in type 2 diabetes(T2DM) and normal glucose tolerance(NGT) first degeree relatives of T2DM.Methods (1) We studied 553 subjects from Chongqing Han families which were enrolled . 251 of them were T2DMS, 175 of them were the NGT first degree relatives(FDR) of type 2 diabetes of the families, and 127 of them were the spouses of T2DM family members. The Anthropometry parameter, blood pressure, renal function parameter, oral glucose tolerance test(OGTT), lipid levels, high sensitive C-reactive protein(hsCRP) were examined. IGR were defined according to World Health Organization (WHO) diagnostic criteria. (2) The Gly15Gly variation(T>G) in exon 2 of adiponectin gene was genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) assay in the subjects. The clinical and biochemical indicators were compared among the genotypes between the groups. The SPSS statistics software was applied to analyse the relations between hs-CRP and the gene polymorphism. Results (1)The distribution of TG+GG genotypes and G allele frequency of adiponectin gene Gly15Gly variation(T>G) in exon 2 were higher in T2DM group than those in the normal control group; There was no obvious distinction of the distribution of genotypes and allele frequency between the normal control group and the NGT FDR group. (2)In T2DM group, the CRP level and abdominal circumference(AC) of TG+GG genotype. were higher than those of TT genotype(P<0.05). In IGR FDR group and normal control group, clinical and biochemical indicators and hs-CRP levels of TG+GG,TT genotype had no significant deviation(P>0.05). (3)Logistic analysis showed that CRP levels were correlated with TG+GG genotype in T2DM group(P<0.05); while in FDR and normal control groups, CRP levels had no correlation with TT and TG+GG genotype(P>0.05).Conclusion Adiponectin gene Gly15Gly variation(T>G) in exon 2 was correlated with CRP in T2DM of the T2DM families. There was no association between the variation and the NGT FDR. The variation of adiponectin gene maybe had influence on the inflammatory reactive and obese in T2DM.
