PART 1 The expression of mediators of inflammation in the rat model for nonbacterial chronic prostatitisObjection: To study the expression of transforming growth factor-beta(1)(TGF-β1),connective tissue growth factor(CTGF), interleukin-1β(IL-1β) and Tumor Necrosis Factor-α(TNF-α) in the prostate tissue of rat model for nonbacterial chronic prostatitis and the mechanism of nonbacterial chronic prostatitis and its fibrosis,to prevent beforehand its fibrosis.Method: 30 10-month-old male Sprague-Dawley rats were divided into two groups randomly,the control group and the model group.The control group received no treatment.The model group had been castrated and injected estradiol benzoate (0.25 mg/kg/day, s.c.) for 30 days.The control group were treated with sham operation and placebo.After the modeling had been done,the prostates of the two groups were taken.The Pathologic appearance of the prostate tissue of the two groups were observed with HE staining and light microscope.The expression of TGF-β1,CTGF,IL-1βand TNF-αof the two groups were detected with immunohistochemical method.Results: The control group did not have inflammatory reaction in their prostates,while the model group displayed visible inflammatory reaction. There is negative expression of TGF-β1,CTGF,IL-1βand TNF-αin control group and strongly positive expression of TGF-β1,CTGF,IL-1βand TNF-αin the model group.There was significant difference of the expression of TGF-β1,CTGF,IL-1βand TNF-αbetween the model group and the control group,according to the result of photodensitometry and area density analysis(P<0.01).Conclusion: TGF-β1,CTGF,IL-1βand TNF-αplay an important role in the development of the nonbacterial chronic prostatitis.TGF-β1,CTGF may be related with fibrosis of parastata tissue in later stage of nonbacterial chronic prostatitis.TGF-β1,CTGF,IL-1βand TNF-αmaybe become a new detection index in CNP and its fibrosis.PART 2 The effects of sinomenine in the rat model for nonbacterial chronic prostatitisObjection: In our present research we adapted nonbacterial chronic prostatitis model to study the effection of sinomenine in nonbacterial chronic prostatitis,and to investigate the possible mechanism of CNP and its fibrosis,provide a new method in clinical treatment and precaution of CNP and its fibrosis.Method: 45 10-month-old male Sprague-Dawley rats were divided into three groups randomly,the control group,the model group and the intervention group.The model group and the intervention group had been castrated and injected with estradiol benzoate (0.25 mg/kg/day, s.c.) for 30 days,and the control group were operated shamly and injected with physiologic saline.After the model finished,the intervention group was given SIN 30mg/kg/d,po,and others were given the same physiologic saline,po. After treatment for 4 weeks,the number of WBC and lecithin body in prostate tissue were measured,the pathologic appearance of the prostate tissue of the three groups were observed by HE staining and light microscope.TGF-β1,CTGF,IL-1βand TNF-αin prostate were detected by means of immunohistochemictry(SP).Then TGF-β1 mRNA were detected by means of RT-PCR.Results: The control group did not have inflammatory reaction in their prostates.The model group displayed visible inflammatory reaction. Inflammatory reaction remitted obviously in the intervention group.There is negative expression of TGF-β1,CTGF,IL-1βand TNF-αin normal group,strongly positive expression of TGF-β1,IL-1βand TNF-αin cellula epithelialis of the model group,strongly positive expression of CTGF in cellula epithelialis and cellula intersitialis of the model group,weakly positive expression of TGF-β1,IL-1βand TNF-αin cellula epithelialis of the intervention group,weakly positive expression of CTGF in cellula epithelialis and cellula intersitialis of the intervention group.The expression of TGF-β1,CTGF,IL-1βand TNF-αin the control group became significantly lower than the others group(P<0.01).The expression of TGF-β1,CTGF,IL-1βand TNF-αin the intervention group were lower than that of in the model group(P<0.01).Conclusion: SIN can remarkably low down the expression of TGF-β1,CTGF,IL-1βand TNF-αin the rat model of nonbacterial chronic prostatitis and maybe a new treatment for the nonbacterial chronic prostatitis,a new precaution for the nonbacterial chronic prostatitis and its fibrosis.
