Study Of Relationship Of The Levels Of Serum SICAM-1, SVCAM-1 And The Amount Of White Blood Cell And Acute Cerebral Infarction

Objective: To study the action of serum soluble intercellular adhesion molecule-1 (sICAM-1),soluble vascular cell adhesion molecule-1(sVCAM-1) and white blood cell(WBC) in onset and development of acute cerebral infarction,to explore the correlation between them and the cerebral infarct volume,the extent of neurological deficits,and to provide important experimental evidences and new methods for therapy and prevention in cerebral infarction.Methods: The levels of serum sICAM-1 and sVCAM-1 were measured serially by two-layer antibody sandwich enzyme-linked immunosorbent assay (ELISA),and the amounts of WBC and neutrophil were taken count with the Beckman in 56 acute cerebral infarction,30 recovered cerebral infarction and 31 age-matched healthy control subjects.Results:1. The levels of serum sICAM-1 and sVCAM-1 in acute cerebral infarction group, recovered cerebral infarction group and healthy control group were significantly different(P<0.01).Furthermore,the levels of serum sICAM-1 and sVCAM-1 in acute cerebral infarction group were markedly higher than those of recovered cerebral infarction group(P<0.05),the levels of serum sICAM-1 and sVCAM-1 in recovered cerebral infarction group were higher than those of healthy control group(P<0.05).2. The amounts of WBC and neutrophil in acute cerebral infarction group were significantly higher than those in recovered cerebral infarction group and healthy control group(P<0.01),but there was no significant difference between recovered cerebral infarction group and healthy control group(P>0.05).3. The levels of serum sICAM-1 and sVCAM-1 in big infarct volume group,middle infarct volume group and small infarct volume group were not significantly different(P>0.05).4. There were positive correlations between infarct volume,the extent of neurological deficits and the amounts of WBC, neutrophil in patients with acute cerebral infarction group(P<0.01); but the levels of serum sICAM-1 and sVCAM-1 were not correlated with infarct volume and the extent of neurological deficits in acute cerebral infarction group(P>0.05).5. The positive correlation was found between the level of serum sICAM-1 and the amount of WBC in patients with recovered cerebral infarction group (P<0.05), but the level of serum sICAM-1 was not correlated with the amounts of WBC and neutrophil in acute cerebral infarction group and healthy control group(P>0.05),and the level of serum sICAM-1 was not correlated with the amount of neutrophil in recovered cerebral infarction group(P>0.05).6. The level of serum sVCAM-1 was correlated with the amount of neutrophil in patients with acute cerebral infarction group(P<0.05),and there was positive correlation between the level of serum sVCAM-1 and the amounts of WBC and neutrophil in recovered cerebral infarction group(P<0.05);the level of serum sVCAM-1 was not correlated with the amount of WBC in acute cerebral infarction group(P>0.05),and no remarkable correlation was found between the level of serum sVCAM-1 and the amounts of WBC and neutrophil in healthy control group(P>0.05).Conclusions:1. The levels of serum sICAM-1 and sVCAM-1,and the amounts of WBC and neutrophil in acute cerebral infarction group were significantly higher than those in other groups,which suggested that sICAM-1,sVCAM-1 and WBC were involved in the pathogenesis of acute cerebral infarction in humans.2. The levels of serum sICAM-1 and sVCAM-1 in patients with acute cerebral infarction were not correlated with infarct volume and the extent of neurological deficits.3. The amounts of WBC and neutrophil in patients with acute cerebral infarction were positively correlated with the extent of neurological deficits and infarct volume, which suggested that WBC might play a role in onset and development of acute cerebral infarction.4. The levels of serum sICAM-1 and sVCAM-1 were remarkably correlated with the amounts of WBC and neutrophil in patients with cerebral infarction,which suggested that sICAM-1and sVCAM-1 mediated adhesive attraction between WBC, neutrophil and endothelium cell(EC),and promote onset and development of cerebral ischemia in humans.
