Chuanxiong, the dried rhizome of Ligusticum chuanxiong Hort (Umbelliferae), is one of the major clinically used cardiovascular-protective botanic medicines in China. Having a reputation for facilitating blood circulation and dispersing blood stasis, this herb is commonly prescribed for the treatment of angina pectoris, cardiac arrhythmias, hypertension and stroke.Lactone compounds, with the same nuclear structure as that of phthalide, are the important active components of Ligusticum chuanxiong Hort. Recent researches indicate that lactone components in Ligusticum chuanxiong Hort can improve the cerebral blood flow through inhibiting thrombus formation and platelet aggregation during ischemic attack, and have exhibited promising effects that are used as preventive and therapeutic agents for antiatherosclerosis. But up to date the commercial reference of lactones in Chuanxiong have not been available. In this thesis, column chromatography was used to isolate them. The High Performance Liquid Chromatography peak area normalization method was used to examine the purity. Their purity were greater than 98%,and MS, IR, UV,1 H-NMR were set up to identify the structure. At last we got reference of ferulic acid, S-I, S-H, S, L. Currently, the quality assessment standard of Chuanxiong is based on the content of the marker compounds ferulic acid or/and TMP. However, due to multiple constituents that might be involved in the therapeutic functions, the content of a single maker compounds can not accurately reflect the quality of Chuanxiong products. In this thesis, the HPLC method for determination of thesis major constituents in the Chuanxiong was built up. The condition was :150mm×4.6mm i.d,ODS-C18 column, methanol: water: acetic acid,the flow rate 0.8ml/min, detection wavelength 280nm,column temperature 30°C and 0.01 AUFS .We calculated the content of the six major constituents in the samples using external standard calibration method. The method of determination developed and validated in the thesis was demonstrated to be convenient, accurate and good reproducibility which was suitable for inspecting the quality of Chuanxiong herb and Chuanxiong-containing products.
