The prevalence of oral lichen planus(OLP)ranks second in oral mucosal diseases.Partial cases can transform from OLP into cancer.Accordingly,the World Health Organization(WHO)presume this disease a possible precancer.The etiology and pathogenesis are still obscure.There are extensive data to suggest that OLP is a T cell-mediated immune damage.Much effort has been spent on OLP therapy,but the outcome is still far from satisfaction.Histopathologically,OLP is characterized by subepithelial band-like inflammatory infiltrate,variable numbers of intraepithelial mononuclear cells focused to the basal keratinocytes and degeneration of basal cells. Previous reports revealed that regulated upon activation normal T cell expressed and secreted factor(RANTES)both can induce lymphocyte to activation and regulate inflammation.Accordingly,we presume that RANTES may act a significant role in the invasive course of OLP.Objective:To explore the role of RANTES and CCR3 in OLP,we assess the expression of RANTES and CCR3 in OLP and normal oral mucosa at the level of mRNA and protein by ISH and immunohistochemistry respectively.Methods:1.OLP(n=30)and normal control(n=10)tissues were defined clinically and pathologically.Seven serial sections were cut for HE staining,immunohistochemical staining,ISH and negative control(3 sections)respectively.We observed the expression of RANTES and CCR3 in different tissues at the levels of mRNA and protein using ISH and IH respectively.2.Digital image processing was used to analysis the expression of RANTES, CCR3 and RANTES mRNA.The optical density(OD)of the different tissue sections was obtained by the means of semi-quantitation.3.Data were analyzed by ANOVA and Student-t test,and expressed as the means(?)±s.Values of P<0.05 were considered statistically significant.Results:1.IH results of RANTES:In normal mucosa,RANTES was positive in stratum spinosum,stratum granulosum and stratum basale with a weak staining,and was identical in some lymphocytes,vascular endothelial cells and fibroblasts iof connective tissues.In OLP,RANTES was intensively positive in stratum spinosum and stratum granulosum;RANTES could also be seen with a medium staining in some lymphocytes and vascular endothelial cells in connective tissues.2.IH results of CCR3:In normal mucosa,CCR3 was positive in stratum spinosum and stratum basale with a weak staining,and was identical in partial lymphocytes of connective tissues.In OLP,CCR3 was intensively positive in stratum spinosum and stratum granulosum;in connective tissue,CCR3 could also be seen in mass sporadic lymphocytes with an intensive staining and in vascular endothelial cells with a medium staining.3.ISH results:In normal mucosa,RANTES mRNA had weak positive expression in stratum basale and the epithelial cell nearby stratum basale and could not be seen in stratum corneum and superficial epithelial cell;in connective tissue, RANTES mRNA had weak positive expression in partial lymphocytes and vascular endothelial cells.In OLP,R.ANTES mRNA had dominated positive expression in stratum spinosum and stratum granulosum;in connective tissue,RANTES mRNA had dominated positive expression in partial lymphocyte and vascular endothelial cell.4.Results ofOD:RANTES,CCR3 and RANTES mRNA in OLP had a significantly higher(P<0.05)OD values(P<0.05)than the normal control.Conclusion:The augmentation and distribution difference of RANTES and CCR3 expression in OLP suggested RANTES may play an important role in OLP development and progression.RANTES may be useful for providing an original therapy of OLP.
