ObjectiveTo explore the effects of Liuwei Dihuang PilI(LW)on learning and memory, serum insulin,blood glucose and morphologic change of pancreatic gland in dementia mice model with diabetes mellitus,To discuss the mechanism of Liuwei Dihuang PilI(LW)intervention to diabetes mellitus and dementia.MethodOne hundred 18-22g KM mice were assigned to the Diabetes group(D),Compound model group(C),Diabetes intervention group(DI),Compound model intervention group(CI)and normal control group by block randomized distribution.Compound model intervention group(CI)were injected intraperitoneally with streptozotocin(40mg/kg)for 5 consecutive days after they were gavaged with Liuwei Dihuang PilI(LW)for 9 consecutive days followed by injections of Scopolamine(1mg/kg)half an hour before the training of Morris water maze test.DI were injected.with normal saline while CI were injected with Scopolamine.D and C were garaged with normal saline while CI and DI were gavaged with Liuwei Dihuang Pill(LW).The normal control group were injected with normal saline and gavaged with normal saline After 9-day treatment with Liuwei Dihuang PilI(LW),5-day streptozotocin. injection followed.These mice were trained by Morris water maze test form 25 to 31 days.After finishing 5 days' Morris water maze test,we killed all the mice,collected their blood by excising the eyeballs and separated the pancreaty gland.All the samples from mouse model were samplinged and assayed. The data were analysis with SPSS11.5,using one-way ANOVA,rank sum test, repeated measures model respectively.Results 1.The Morris water maze test includes two parts:Place Navigation Training and the Spatial Probe Test.1.1 Position navigation test results:1.1.1 Comparisons between the five groups:Compared with the normal group, the compound model group has a significant difference(P<0.05),suggesting that the learning and memory capacity of model decreased significantly.1.1.2 Trend of the five-day changes:The mean of escape latency,distance become smaller gradually with the training number increasing,suggesting that learning and memory capacity of mice is improved after the training.1.1.3 Search strategy:Compared with the normal group,the percentage of straight line type and trend type in compound model group are decreased significantly(P<0.05),suggested that learning and memory capacity in compound model group are decreased significantly.Compared with the compound model group,the percentage of straight line type and trend type in compound model intervention group was increased significantly(P<0.05).1.1.4 Comparisons the mean escape latencies among the five groups:as the time go on,the mouse learned to find platform gradually under the training. The graph show thatthe slope rate of Liuwei Dihuang Pill(LW)treatment group is far larger then other group,suggesting that the progress of Searching platform in Liuwei Dihuang Pill(LW)treatment group is more faster.1.2 Spatial Probe test results:The time in the fourth quadrant and the cross-platform number showed that the differences were no statistically significant(P>0.05)among five groups.Based on the above results,scopolamine harmed the ability of learning and memory in mice,modeling success.Liuwei Dihuang Pill(LW)to some extent improved the mice's learning and memory capacity,mainly in Search strategy while no significant improve in other Indicators.2.The effect on blood glucose.2.1 Blood glucose levels comparison beween five groups after modeling:Compared with the normal group,the diabetes group,the compound model group have significant difference in blood glucose levels(P<0.05);Compared with diabetes group,the blood glucose levels of diabetes intervention group are lower,but it was no significant(P>0.05);Compared with compound model group,the blood glucose levels of compound model intervention group are lower,but it was no significant(P>0.05).This shows modeling success,the blood glucose levels of model group had increased after STZ injection.2.2 Comparison after modeling in different time.With the passage of time,blood glucose increased gradually.There are no statistically significant differences between the 3rd time and 4th time measurement of blood glucose levels(P>0.05).2.3 Comparison among five groups in different time.Blood glucose level in normal group change not significantly in different times while diabetes groupand Compound model group tended to increase significantly.Compared with diabetes group,diabetes intervention group tended to increase moregently,suggesting that blood glucose level increase trend was slowed under the intervention of traditional Chinese medicine.3.The effect on serum insulin.Compared with normal group,then serum insulin levels in diabetes group and compound model group are tend to decrease(P>0.05).Compared with diabetes group,then serum insulin levels in diabetes intervention group are tend to increase(P>0.05).4.The protection effect on islet cells.Compared with normal group,then islet cells in diabetes group and compound model group are damaged more seriously(P<0.05);Compared with compound model group,then islet cells in compound model intervention group are damaged more gently(P<0.05),suggesting that Liuwei Dihuang PilI(LW)can protect the islet cells damaged by STZ.Conclusion1.The escape latency and distance in the dementia mice model with diabetes mellitus are longer than in the normal group,which suggests the impairment in the capacity of learning and memory.2.Liuwei Dihuang PilI(LW)can improve the learning and memory capacity of mouse harmed by scopolamine,mainly in the angle of Search strategy.3.Liuwei Dihuang Pill(LW)to some extent protected the islet cells of mice damaged by STZ,but there is no significant improve in serum insulin and blood glucose level.
