ObjectiveDepression is a very serious psychosis disorder the world nowadays.With the development of the society and the disease,its affect to people's health becomes distinct day by day.According to the report of WHO,the among all kinds of diseases,and the financial burden caused due to depression totok the fourth place globally.The quota index was expected to jump to the second rank place in 2020.As a result,it becomes urgent for medical and scientific workers experts to search for effective treatment,and clarify its function mechanism.The pathologic mechanism of depression has not been completely revealed scientifically.The monoamine neurotransimtter theory took the leading place in the past.Now general point of view is that psychology is very important to depression.Another,with further research,experts in this area posed advanced several new theories,such as theory of damage to the nurtrition of cerebral nerves.BDNF and TGF-β1 are important parts in this new theory.Music therapy and acupuncture are important parts in the therapy of depression.They have a lot of relations,the ancients have recognized this and many studies have proved that in the recent years.Music electrical acupuncture is based on the electro-acupuncture and music therapy,This can,to some extent,alleviate the symptoms of patients suffered depression,with its advantage of easy praeting,and without side-effects,it can adjust psychology and physiology,though the mechanism of its effect remains unclear,our research focused on observing the effect of music electrical acupuncture on changes of behavior in depression rats,furthermore,exploring some possible mechanism.s by investigating the influence of music electrical acupuncture on BDNF,c-fos,TGF-β1 expression in rat hippocampus.MethodsForty male rats were evenly randomized into control,model and music electrical acupuncture groups.Depression model was established by using chronic unpredictable mild stress(CUMS)stimulation(forced ice-water swimming,electie shock,tail-clamping,etc) combined with lonely raising for 21 days.Music electrical acupuncture was applied to Baihui(GV 20)and Sanyinjiao(SP6),once daily,continuously for 21 days,The rat's body weight,open-field test were recorded on the 1thand 21stday during experiments respectively.After the behavior observation,BDNF expression changes ofhippocampus in music electrical acupuncture intervened depression model rats were observed.ResultClinical study-both music electrical acupuncture and Fluoxetine were effective in the treatment of major depression.For relieving endogenous depression,Fluoxetine was better.The score on the syndrome of anxiety somatization(SAS)were significantly lower in music electrical acupuncture group than the Fluoxetine group.The score on Asberg Rating Scale of Fluoxetine group was significantly higher than music electrical acupuncture group.Since Fhoxetine has more side effcts than music electrical acupuncture, therefore,the efficacy index of music electrical acupuncture group is significantly higher than Fluoxetine group.Experiment study-compared with contol group,the weight increase was slow and the activities of rats in open-field test in model group were significantly decreased.Music electrical acupuncture can improve behavioral disorder of depression rats. Music electrical acupuncture can increase the levels of monoamine neurotransmitter in central nervous system.Immuno-histochemistry results showed that the number of Brain-derived neurotrophic factor(BDNF)positive neurons in hippocampus of depressed rats in model group was much more than the control group.Music electricai acupuncture can increase the number and improve the appearance of BDNF neurons in the depressed rats,but decrease the number and influence the appearance of c-fos neurons in the depressed rats.The TGF-β1 of depressed rats in model group were significantly increased,Which was significantly decreased in the music electrical acupuncture group.Conclusion1.Music electrical acupuncture at "Baihui(GV 20)and Sanyinjiao(SP6)could prevent or suppress the onset of depreession and raise the volume of srcrose -intake in depression rats.2.Music electrical acupuncture at "Bai hui"(GV 20)and Sanyinjiao(SP6)could increase the expression of BDNF mRNA in hippoeampus of depression rat,thus protect the neuron in hippocampus.3.The TGF-β1 of depressed rats in the the music electrical acupuncture group was significantly decreased,thus protect the neuron in hippocampus.
