Objective:To observe the effects on depression model rats diet, body weight and behavior of music electroacupuncture science, expression changes through the hippocampus in rats were examined by EAAT3and GAD65proteins, to explore the mechanism of music electroacupuncture anti depression.methods:The SPF level of40SD rats, male and female, body weight180-220g, do behavior score by open-field method. In7:00-19:00light,19:00-7:00dark lab, after1weeks of feeding,40rats were numbered, were randomly divided into control group, model group, fluoxetine group and the music electroacupuncture group, with10rats in each group. The control group of5per cage, the remaining4per cage1. By chronic unpredictable mild stress model of depression:from the group of first to twenty-first days, except the blank control group, the other groups animal underwent different stress, including foot shock (current intensity of1.0mA, every1minutes to stimulate the1times, each time for10seconds, a total of30times, swimming in cold water(4.0℃,5minutes), shaking (1times/sec,15minutes), tail clamp (1minutes), bound (3hours), fasting (24h), fasting (48h) and turned night into day, every day of randomly arranged a stress, average each stimulus used2times. Music electroacupuncture group was given acupuncture Sishencong, Neiguan and Sanyinjiao, fluoxetine group were given fluoxetine1.8mg/kg body weight, dissolved in pure water in the stomach2ml,1times/day. Changes in experiment first days, seventh days, fourteenth days and twenty-first days to observe the rat diet, body weight, behavior, was detected in the hippocampus of rat brain EAAT3and GAD65protein expression after the experiment. The experimental results by Excel2003and SPSS13.0statistical software for analysis and processing.Results:1. In the animal model, each animal weight, sucrose consumption and feed consumption were not significant difference (P>0.05), along with the extension of model making, model group rats weight with slow growth, but compared with the normal group, the rate of increase slowed significantly, compared with two groups, there was significant differences (P<0.05), followed by consumption and feed the rats in the model group for sucrose consumption decreased significantly, compared with the normal group, the difference was statistically significant (P<0.05), Open-Field behavior results in the depression model rats changes indicate that, compared with the normal group, model group animals, horizontal movement and vertical movement of the number of times significantly reduced, significant difference (P<0.05), music EA group, treatment group and fluoxetine group of horizontal motion and vertical motion score compared with the model group, there were significant (P<0.01-0.05), and compared with the normal group, there was no significant difference (P>0.05).2. Expression of EAAT3in hippocampus of model group decreased protein compared with the blank group, while the expression of music electroacupuncture group EAAT3hippocampus protein was significantly increased in model group, fluoxetine hippocampus expression had no obvious change.3. Expression of GAD65in hippocampus of model group decreased protein compared with the blank group, while the expression of music electroacupuncture group GAD65hippocampus protein was significantly increased in model group.Conclusions:1. Music electroacupuncture can effectively increase the depression rats weight, sucrose consumption and feed consumption, stimulate the movement ability.2. Music electroacupuncture can regulate the expression of EAAT3in the hippocampus of rats with depression increased protein, glutamate neurotoxicity of conversion, reduction of excitatory amino acid, increase the expression of hippocampal GAD65protein, improve the symptoms of depression, which may be one of its mechanisms of action.
