model-based chemotherapeuty treatment design and protein profiling of serum with SELDI-TOF-MS in pancreatic cancerBackground and Aim:Pancreatic cancer spreads very fast,and it is difficult to early diagnose.CA19-9 is prevalently used as the best serum tumor marker.Recently,the development of proteomics provides new sight in searching for new tumor markers.We intended to establish human pancreatic cancer model from subcutane to orthotopic transplantion with panc-1 in athymic mice,and construct the chemotherapeutic treatment model of pancreatic cancer.To detect the serum proteomic patterns in Pancreatic cancer patients by using proteomics technology,screen biomarker candidates,build diagnostic models and evaluate its clinical significancel.Methods:Athymic mice are divided into three groups at random:the control group,the pancreatic cancer group and the gemcitabine group.We detect the serum proteomic patterns with the SELDI-TOF-MS technology and protein chips,analyze the original data and screen the serum markers in the way of informatics,to conduct a diagnostic model as well as an evaluation system for chemotherapy.Results:The cancer inhibition rate of gemcitabine group is 50.14%.The modelâ… composed by 3 protein peaks(11753,8615,8256m/z)could do the best in the diagnosis between pancreatic cancer and normal.Its sensitivity and specificity were 95%(19/20) and 95%(19/20).The modelâ…¡formed by 2 protein peaks(3879,4772m/z)could do the best in the diagnosis between pancreatic cancer and gemcitabine group.Its sensitivity and specificity were 100%(19/19)and 95.24%(20/21).Conclusions:The method showed great potential for the early detection,staging before operation and screening novel and better biomarkers for pancreatic cancer.
