Objective: To localize glycodelin and test its expression in deciduas andmeasure it in serum from normal pregnancy, mild preeclamptic and severe preeclamptic patients and then, to discuss the role of glycodelin in the onset of this disease.Materials and Methods: Deciduas and serum were obtained from 33 normalpregnant women, among them 8 were mild preeclamptic patients and 24 severe preeclamptic patients who had caesarean section to stop pregnancy. Preeclampsia was diagnosed and classified according to strict criteria recommended by the 8th edition of Gynecology and Obstetrics edited by Youji Feng and Qian Shen. Exclusion criteria were multiple gestation, gestational complications, active labor and signs of other cocurrent medical complication. The subjects of control had no signs of gestational complications and singleton pregnancy. Immunohistochemistry was used to locate glycodelin in various tissues. Western blotting and enzyme linked immunosorbent assay were employed to detect glycodelin protein in decidua and serum. Data were normally distributed as tested by the Kolmogorov-Smirnov test and presented in means and standard errors of mean. Statistical analysis was performed by employing the one-way ANOVA or Student t test where appropriate. P value less than 0.05 was considered statistically significant. Results (1) Glycodelin has been localized by immunohistochemical staining tosecretory endometrial glands, the glandular epithelium of deciduas, and the deciduas cells. (2)There is no difference in the expression of glycodelin in deciduas between patients and control.(3)Compared with control group, glycodelin is significantly lower in maternal serum than that in preeclampsia group. It is worth noting that lycodelin in severe preeclampsia group significantly lower than control.Conclusions:1. Glycodelin is mainly localized in secrete phase endometrium, epithelium of pregnancy decidua glands and decidua cells. There is no difference of the expression of glycodelin in decidua between preeclamptic group and control group.2. Compared with control group, glycodelin is lower in maternal serum of preeclamptic group. Furthermore, it is significantly lower in the severe preeclamptic group than control. The decreased serum glycodelin that leads to the excessive activation of maternal immune system may be one of etiological factors of this desease.
