Objective: To study the optimal extraction technology, primary methods of seperation and purification of naringin from pomelo pericarp with ethanol. To observe naringin's hypoglycemic activities in diabetic animal model induced by different reagent.Methods: A HPLC method was established for the determination of naringin. The method of orthogonal design was used to determine the effect of the ethanol's concentration , extraction temperature,extraction time and quantity of solvent on the extraction yield of naringin.Naringin was isolated by column chromatography of polymide and purified by two recrystallization. The diabetic mouse model was established by injecting alloxan into abdominal cavity. Blood glucose,activity of SOD, concentration of MDA in serum and liver of the diabetic mice were determined .The type 2 diabetic rat model was established by both high lipid forage and injecting low dose STZ into abdominal cavity. FBG, FINS, OGTT, TC,TG,activity of SOD and concentration of MDA were measured.Results: We establish a HPLC method for determination of naringin. The optimal extraction parameters were: ratio of solid to liquid as 1:20, 60% ethanol, temperature 90℃and 2 hours.After two recrystallization,the purity of naringin was 75.61% and its recovery rate was 19.38.Naringin can greatly decrease FBG level, MDA content and increase the activity of SOD in mice. In addition , the levels of FBG,FINS,TC,TG,OGTT in serum and the content of MDA in liver tissue were reduced and the activity of SOD in liver tissue was increased significantly in rats.Conclusion: In the optimal extraction and isolated condition , the purity of naringin is modestly high.However,a further study is required to improve the purity. Naringin can increase the antioxidant level in mouse and rat,correct disorders of glucose and lipid metabolism and reduce the resistance of insulin.
