Hypertrophic scarring is the major significant negative outcome after survival from of a thermal injury,especially in deep burned patients.Excessive proliferating fibroblasts and overabundant wound matrix are responsible for raised,red and inflexible scar tissue,which causes itching and pain,and might lead to serious functional and cosmetic problems for the patients.The mechanism of its formation is still in debating,by now we reach consensus on it, the fibroblasts play an important role in the process.Over the years,we keep on searching for a reliable surgical excision,intralesional corticosteroids injection, radiation,laser surgery,cryotherapy and other medication,combined ways can get better results than the sole one.But unfortunately We have not yet been successful in preventing and treating hypertrophic scar.So to seek the effective way and medicine for prevention and cure hypertrophic scar is a very important topic on plastic surgery science.To provide experimental bases for clinical treatment of hypertrophic scar,we do this study.Objective:to study the effect of hirudin and tanshinone on hypertrophic scar fibroblasts' proliferation and secretion,and provide theoretical evidence for clinical medication.Methods:the hypertrophic scar fibroblasts were from 10(male:6,female:4) persons' hypertrophic scar samples.We investigated the effect of hirudin and tanshinone on HSFB with exposure to 0 u/ml,1u/ml,10u/ml or 50u/ml hirudin or 0ug/ml,1ug/ml,10ug/ml or 100ug/ml tanshinone for 24,48 or 72hours in vitro model.We observed cells' appearance change by inverted phase contrast microscope,detected the cells' vigor and proliferation by MTT method, measured the concentration of TGF-β1 by enzyme-linked immunosorbent assay, determined the gels' contractile index and assessed the expression ofⅠ,Ⅲprocollagen mRNA by reverse transcription PCR.Results:hirudin and tanshinone could inhibit fibroblasts' proliferation and vigor,reduced fibroblasts' secretion of TGF-β1,decreased the contractile index of FPCL and diminished the expression ofⅠ,Ⅲprocollagen mRNA. Tanshinone's action was strengthened with the increasing of its concentration.Conclusions:the experiment proved hirudin and tanshinone has a distinguished inhibitory action to proliferation,secretion and contraction of HSFB in vitro.Compared with plane cultural model,FPCL has slower and weaker reaction of medicine.FPCL is more vivid model of human hypertrophic scar.
