Quality problems of Schisandra chinensis(Turcz.)Baill(Schisandra chinensis)and Schisandra sphenanthera Rehd.et Wils.(Schisandra sphenanthera)are increasing owing to their obvious difference of supply and demand.At present,there are only quality standards of materials without the quality control of their extract for Schisandra chinensis and Schisandra sphenanthera in Chinese Pharmacopoeia.And a single indicator for quality control could not really reflect their intrinsic quality.In this paper,qualitative and quantitative studies were carried out on the contents of various components(including Schisandrol A,Schisandrol B,Schisantherin A,Schisantherin B,Schisandrin A,Schisandrin B and Schisandrin C)in Schisandra chinensis,Schisandra sphenanthera and their extracts.In addition,the established quality standards of materials and extracts and the devoloped fingerprints provided the reference for the quality control of the raw materials and extracts of Schisandra chinensis and Schisandra sphenanthera.In this study,a UHPLC analysis method for the rapid analysis of seven lignans in14 min was established.This method showed a average recovery 98.29%-102.50%with RSD 1.37%-1.86% for Schisandra chinensis,and a recovery 99.08%-100.53%with RSD 1.11%-1.73% for Schisandra sphenanthera,which proved that this method was rapid,stable and reliable.The contents of 10 batches of Schisandra chinensis and9 batches of Schisandra sphenanthera were determined by this method.The results showed that the contents of lignans in Schisandra chinensis and Schisandra sphenanthera had significant difference between different origins and batches.It was suggested that the quality standard of Schisantherin B and Schisandrin A(mass fraction: not less than 0.08% and 0.40%,respectively)for Schisandra sphenanthera and Schisandrol B,Schisantherin A,Schisantherin B,Schisandrin A,Schisandrin B and Schisandrin C(mass fraction: not less than 0.10%,0.01%,0.05%,0.04%,0.20%and 0.04%,respectively)for Schisandra chinensis should be set up.Based on the mapof c Schisandra sphenanthera(Shanxi)and h Schisandra chinensis(Liaoning N3)as reference map,the fingerprints of Schisandra chinensis and Schisandra sphenanthera were respectively established by using the National Pharmacopoeia Commission "Chinese Journal of Chromatographic Fingerprint Similarity Evaluation System(2004A)" for data analysis.The similarity of Schisandra chinensis was above 0.991,and the similarity of Schisandra sphenanthera was above 0.976(except for the m Schisandra sphenanthera(Gansu)).Taking the control map of Schisandra sphenanthera as the reference map,the fingerprints of 9 batches of Schisandra sphenanthera and 10 batches of Schisandra chinensis were analyzed.The fingerprints of two materials were different from each other,from which two materials could be distinguished accurately and easily though the similarity.In this paper,through single factor experiment and response surface method,the effect of material ratio,extraction time and ethanol concentration on lignans extraction through ethanol reflux and ultrasonic extraction were investigated.The optimal extraction technology of lignans in Schisandra chinensis was as follows:material ratio 1: 9,extraction time 2 h,ethanol concentration 90%,extraction temperature 90 ? and extraction times 2 times.The results showed that the macroporous resin was AB-8.The effect of sample volume,adsorption influencing factors,the consumption of purified water used for elution and the concentration and usage of ethanol on lignans adsorption were determined by dynamic adsorption experiment.The optimal purified technology of lignans in Schisandra chinensis was as follows: sample volume 2 BV,loading speed 3 BV/h,purified water 6 BV and80% ethanol 5 BV.In this paper,the corresponding components of standard extract for Schisandra chinensis were determined by lignans determination method of medicinal material.The results showed that the content of lignans in the standard extract was higher than that of the material,which indicated that the preparation process of lignans had a goodenrichment effect for Schisandra chinensis.Through the study on the peak of Schisandra chinensis and its corresponding extract,each peak could be corresponded one by one,and no index component was missing,which meaned that the quality of the extract could be controlled from the material.And it was suggested the content of Schisandrol A,Schisandrol B,Schisantherin A,Schisantherin B,Schisandrin A,Schisandrin B and Schisandrin C for Schisandra chinensis were not less than 2.0%,2.0%,0.5%,0.10%,0.25%,0.25%,1.0% and 0.2% respectively,and Schisantherin A,Schisantherin B,Schisandrin A for Schisandra sphenanthera were not less than 1.0%,0.4% and 2.0% respectively.
