Objective To assess the association between +299G/A variant of resistin gene,plasma resistin, and abdominal obesity,insulin resistance. Methods The resistin +299G/A polymorphism was identified by polymerase chain reactin restriction fragment length polymorphism (PCR-RFLP) in 125 cases of abdominal obesity and 85 cases of control in Chinese. Their height(H), weight(W) and waist circumference(WC) were measured. Fasting plasma glucose (FPG),Fasting insulin (FINS), fasting plasma resistin(Resistin),total cholesterol(CHO), triglycerides(TG), high density lipoprotein– cholesterol(HDL-c),apolipoprotein A(ApoA) and apolipoprotein B(ApoB) were also measured , HOMA-IR=FPG×FINS/22.5 as insulin resistance indexes was used to analyse the relation between resistin and insulin resistance. The association between +299G/A variant of resistin gene,plasma resistin, and abdominal obesity,insulin resistance were assessed by SPSS13.0.Results 1. There was significant difference of GG,AA genotypes and G/A alleles frequencies between patients with abdominal obesity and health subjects(P<0.05).In patients with abdominal obesity,the plasma resistin and Homa-IR in AA genotype was higher than that in GA genotype, and both were higher than that in GG genotype.2.Resistin, FINS, Homa-IR in abdominal obesity group were significantly higher than those in control group.3. The multiple stepwise regression analysis proved that resistin was positively correlated with FINS, Homa-IR and WC(P﹤0.01).Conclusion The +299G/A of resistin gene polymorphism was associated with susceptibility to abdominal obesity.The allele A has an increased risk of abdominal obesity and insulin resistance.The level of fasting plasma resistin increases in patients with abdominal obesity.The correlation between Homa-IR and resistin suggests that resistin was involved in insulin resistance.
