Objective: To explore whether the polymorphism of single nucleotide polymorphisms (SNPs) of Resistin is associated with the pathogenesis of type 2 diabetes.Methods: Screening for mutations in the entire coding region for the Resistin gene were performed by the polymerase chain reaction-single strand conformation polymorphism(PCR-SSCP) in 349 An Hui Province of han nationality in china. The subject were divided into two groups: 307 cases of type 2 diabetes ,102 cases of control .The study including the distribution of the frequency genotypes and alleles of the polymorphism of Resistin gene among groups , as well as the relation between the polymorphism and some clinical phenotype traits. Meanwhile applying multiple factors step wise regression to know the interation of the polymorphism of Resistin gene on the prevalence of the 2 type diabetes. Results: 1)The ISI is associated with BMI(P=0.007) FBG(P=0.000) PBG(P=0.000) FINS(P=0.000) and PINS(P=0. 005) in Chinese Han population Diabetes of AnHui region .2)The SNP prevalence of Resistin gene polymorphism in Chinese Han population of AnHui region .SNP-157G/A was found in the intron 1;SNP+115 C/G was found in the exon 2;SNP+877 G/C was detected in the intron 3;The SNP3 was conformed to expectation of the Hardy-Weinberg rule. In normal contral, the frequencies of SNP2 G-Alleles was 1. 2%, In diabetic patients, it was 1. 6%, the frequencies of CC were 98. 8%and 98. 2% respectively, the frequencies of CG were 1. 23% and 1. 64% respectively, There were not differ significantly between the groups. In normal contral, the frequencies of SNP3 C-Alleles was 5.8%, In diabetic patients, it was 6.5%, the frequencies of GG were 88.4%and 87.8%respectively, the frequencies of GC were 9. 3% and 11. 4% respectively, the frequencies of CC were 1.16% and 0. 76% respectively, There were not differ significantly between the groups. 3) ISI was assoaited with AGE (P=0. 049, r=-0. 276), FBG (P=0. 003, r=-0. 437), PBG (P=0. 003, r=-0. 444), FINS (P=0. 000, r=-0. 686) and WHR (P=0. 007, r=-0. 399). Conclusions: 1 The SNPs in resistin gene were dected in Chinese Han population of AnHui region using PCR-SSCP. One SNP(-157G/A) was found in intron 1, One SNP(+115C/G) was found in Eexon 2, One SNP(+877G/C) was found in intron 3. 2 The SNP3 was conformed to expectation of the Hardy-Wei nberg rule. The frequency of SNP3 in resistin gene in diabetic patients did not differ significantly from normal control subjects.
