Objective To assess the association between -420C/G variant of resistin gene,plasma resistin, and obesity,type 2 diabetes mellitus.Methods The resistin promoter -420C/G polymorphism was identified by polymerase chain reactin restriction fragment length polymorphism (PCR-RFLP) in 67 cases of obesity ,63 cases of diabetes with obesity and 60 cases of control in Chinese. Their height(H), weight(W) and waist circumference(WC) were measured. Fasting plasma glucose (FPG),Fasting insulin(FINS), fasting plasma resistin(Resistin),total cholesterol(CHO), triglycerides(TG), high density lipoprotein-cholesterol(HDL-c),low density lipoprotein-cholesterol(LDL-c), apolipoprotein A(ApoA) and apolipoprotein B(ApoB) were also measured,HOMA-IR=FPG×FINS/22.5 as insulin resistance index was used to analyse the relation between resistin and insulin resistance. The association between promoter -420C/G variant of resistin gene,plasma resistin, and obesity,insulin resistance were assessed by SPSS13.0.Results 1.Resistin, FINS, Homa-IR in obesity group and obesity with diabetes group,were significantly higher than those in control group(all P <0.05).And there was no difference in obesity and obesity with diabetes group except Homa-IR. 2.The multiple stepwise regression analysis proved that resistin was positively correlated with FINS, Homa-IR and BMI.3.There was significant difference of GG,CC genotypes and G/C alleles frequencies between patients with obesity,obesity with diabetes and health subjects.In patients with obesity and obesity with diabetes,the plasma resistin and Homa-IR in GG genotype was higher than that in GC genotype(both P <0.1), and both were higher than that in CC genotype(all P <0.1). Conclusion Resistin, FINS, Homa-IR in obesity group and obesity with diabetes group,were significantly higher than those in control group.The -420C/G of resistin promoter gene polymorphism was associated with susceptibility to BMI.The allele G has an increased risk of obesity and insulin resistance.The level of fasting plasma resistin increases in patients with obesity.The correlation between Homa-IR and resistin suggests that resistin was involved in insulin resistance.
