Objective: Investigate the effect of Dai Nationality Drugs Clerodendranthus spicatus on adenine-induced CRF rat model,under the guidance of Dai medicine,further improve the principle of Dai Nationality Drugs Clerodendranthus spicatus use,contribute to the clinical use of Chinese national medicines and the development of ethnic medicine,to provide relevant experimental and theoretical basis for clinical prevention and treatment of chronic renal failure.Method: 103 SD rats were selected and weighed 200 ± 20 g,and 6 groups were established.Adenine was used as a model drug to make CRF rat model,15 normal control groups,20 model groups,17 uremic clear granules positive group,Clerodendranthus spicatus high,medium and low dose groups.The daily dose of the stomach was calculated according to the weight of the week.For the model group,the positive group and the Clerodendranthus spicatus high,medium and low dose groups were intragastrically administered with 10 ml/kg adenine(concentration 0.015 g/ml)every morning,and the normal control group was given 10 ml/kg drinking water every morning.In the afternoon,the positive group was given uremic clearing 20ml/kg(concentration 0.3g/ml),and the Clerodendranthus spicatus high,medium and low dose group was given Clerodendranthus spicatus aqueous extract 20ml/kg(concentration 0.6g/ml,0.3g/ml,0.15g/ml),normal control group and model group were given 20ml/kg drinking water.Observe and record the general situation during the experiment.After 30 days of experimentation,fasting for 12 hours,weighed the last weight and recorded,femoral artery blood test biochemical indicators blood urea nitrogen(BUN)and serum creatinine(Scr),dissected observation and weighing the kidney weight,record and The kidney organ coefficient was calculated from the combinedbody weight,and kidney pathological tissue sections were prepared by HE staining.Enzyme-linked immunosorbent assay(ELISA)was used to detect the expression levels of HGF and Bcl-2 in right kidney tissue.The above data was collected and statistical analysis was performed using statistical software.Result:1.General situation:Compared with the normal control group,the rats in the model group showed a decrease in dietary intake with a large amount of drinking water,pale auricles,blind eyes,dry hair,hair loss,fatigue and arch reduction,and decreased activity.Compared with the model group,rats in the Clerodendranthus spicatus high-,medium-dose,and positive groups also had reduced dietary intake and large amounts of drinking water,but the remaining symptoms were significantly improved.The improvement effect of Clerodendranthus spicatus high and medium dosage is better than that of Clerodendranthus spicatus low dose group,suggesting that the general improvement effect increases with the increase of drug concentration.2.weight:Compared with the normal control group,the body weight of the model group decreased,and P < 0.01,the difference was very significant.Compared with the model group,the weight of the positive group was 296.5 ± 23.23 g,P>0.05,and the difference was not statistically significant.Compared with the model group,the body weights of the Clerodendranthus spicatus high,medium and low dose groups were 278.4±24.58 g,286.5±20.91 g,281.9±28.77 g,P>0.05,the difference was not statistically significant.3.Kidney organ coefficient:Compared with the normal control group,the renal organ coefficient value of the model group increased,P<0.01,and the difference was very significant.Compared with the model group,the renal organ coefficient value of the positive group decreased,P<0.01,and the difference was very significant.Compared with the model group,the values of renal organ coefficient in the high,medium and low dose groups of Clerodendranthus spicatus decreased,P<0.01,and the difference was very significant.4.Blood urea nitrogen(BUN)index:Compared with the normal control group,the BUN value of the model group increased,P< 0.01,and the difference was very significant.Compared with the model group,the BUN value of the positive group decreased,P<0.01,and the difference was very significant.Compared with the model group,the BUN value of the high and medium dose groups of Clerodendranthus spicatus decreased,P<0.01,and the difference was very significant.Compared with the model group,the BUN value of the low-dose group of Clerodendranthus spicatus was 26.85 ± 9.741mmol/L,P>0.05,and the difference was not statistically significant.5.Serum creatinine(Scr)index:Compared with the normal control group,the Scr value of the model group increased,P<0.01,and the difference was very significant.Compared with the model group,the Scr value of the positive group decreased,P<0.01,and the difference was very significant.Compared with the model group,the Scr values of the high,medium and low dose groups of Clerodendranthus spicatus decreased,P<0.01,and the difference was very significant.6.Pathological observation of kidney:Dissect and observe the kidney tissue visible to the naked eye.The kidney of the normal control group had normal appearance,the color was reddish brown,the texture was firm and accompanied by good luster,no swelling,the cut surface of the medulla was clear,reddish brown,the medulla was light red,the capsule was shallow,and it was not easy to peel off.The kidney of the model group is gray-brown,the surface is grayish white,the texture is soft and dull,the shape is swollen,the cut surface of the medulla is blurred,the cortex is extremely thin,and there are a large number of crystalline particles in the kidney,and the capsule is easily peeled off.The kidneys of the positive group were reddish-brown and grayish white,with less white particles.The texture was slightly firm,slightly swollen,slightly lustrous,and the cut surface of the medulla was clear.The cortex was close to the thickness of the normal group,and the capsule was tightly bound and not easily peeled off.The kidneys of Clerodendranthus spicatus high and medium dose group were reddish brown and grayish white.The distribution of white particles was less,the texture was softer,the kidneys were slightly swollen,lacking luster,the medulla had a clear cut surface,the cortex was thin,the crystal particles were less distributed,and the capsule was tightly bound.It is not easy to peel off.The renal structure of the normal control group was intact and no abnormalities,and no urate crystals.The renal structure of the model group showed pathological changes,a large number of urate crystals,abnormal structure of the glomerulus,enlarged tubules,partial mild atrophy,a large number of inflammatory cell infiltration,and significant changes in renal fibrosis.Compared with the model group,the renal pathological damage of the rats in the positive group was alleviated,the urate crystals were reduced,and there were a certain amount of inflammatory cell infiltration,renal tubular enlargement,partial mild atrophy,and low degree of renal fibrosis.Compared with the model group,the renal pathological damage of the Clerodendranthus spicatus high-dose group was relieved,the kidney structure was relatively intact,the renal urate crystals were reduced,a small amount of inflammatory cells infiltrated,the structure of the renal corpuscle was clear,and the degree of renal fibrosis was low.Compared with the model group,the renal pathological damage of the rats in the Clerodendranthus spicatus middle dose group was alleviated,the renal urate crystals were reduced,the renal corpuscle structure was clear,the inflammatory cell infiltration was more,and the degree of renal fibrosis was.7.Enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of HGF in kidneys:Compared with the normal control group,the level of HGF in the model group decreased,P < 0.01,the difference was very significant.Compared with the model group,the level of HGF in the kidney of the positive group increased,P<0.01,and the difference was very significant.Compared with the model group,the level of HGF in the high and middle dose groups of Clerodendranthus spicatus increased,P<0.01,and the difference was very significant.8.Enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of Bcl-2in kidneys:Compared with the normal control group,the level of Bcl-2 in the model group decreased,P<0.01,and the difference was very significant.Compared with the model group,the level of Bcl-2 in the kidney of the positive group increased,P<0.05,and the difference was significant.Compared with the model group,the level of Bcl-2 in the high-dose group of Clerodendranthus spicatus increased,P<0.01,the difference was very significant.Compared with the model group,the Bcl-2 level in the kidney of the Clerodendranthus spicatus middle dose group increased,P<0.05,and the difference was significant.Conclusion:Dai Nationality Drugs Clerodendranthus spicatus dose of 6 ~ 12g/kg has a role in reducing the renal organ coefficient,BUN,Scr,improving general condition,protecting kidney function,relieving renal pathological damage and slowing down renal fibrosis in a rat model of CRF induced by adenine.There was no improvement in body weight loss in the adenine-induced CRF rat model.The Dai Nationality Drugs Clerodendranthus spicatus dose of 3g/kg for the adenine-induced CRF rat model has the effect of lowering the renal organ coefficient,Scr,improving the general condition,protecting the renal function,relieving the renal pathological damage,and slowing the renal fibrosis process.However,adenine-induced There was no improvement in BUN elevation and weight loss in the CRF rat model.The effect of Dai Nationality Drugs Clerodendranthus spicatus on adenine-induced CRF rat model may be related to the increase of HGF and Bcl-2 expression in the kidney of CRF rat model and inhibition of renal cell apoptosis and fibroblast proliferation.
