Objective To investigate the genetic polymorphisms of patients with Kawasaki Disease(KD)and that complicated with coronary artery lesion (CAL)in Endothelin-1(ET-1)gene(K198N),Endothelin A receptor (ETAR)gene(70C>G)and Endothelin B receptor(ETBR)gene(G57S).Methods 30 cases of patients with KD and 60 healthy children as controls were studied.Polymorphism analysis of these genes(K198N, 70C>G and G57S respectively)were detected by polymerase chain reaction- restriction fragment length polymorphism method(PCR-RFLP).Results(1)As for the K198N locus of Endothelin-1 gene,there were no significant differences between genotype frequency of GG,GT and TT types in KD and healthy controls(χ~2=0.124,P>0.05);there was no significant difference between allele frequency of G and T types in KD and controls(χ~2=0.053,P>0.05);there were no significant differences between the frequencies of genotype and allele of KD complicated with CAL and without CAL(χ~2= 1.401,0.019 respectively,all P>0.05).(2) For the 70C>G locus of Endothelin A receptor gene,the frequency of G allele in KD is significantly higher than that in controls(χ~2=4.589, P<0.05);the genotype frequencies of GG,CG and CC types in patients with KD was not significantly higher than that in controls(χ~2=5.173, P>0.05);neither the genotype nor the allele frequency was observed significant differences between KD with CAL and without CAL (χ~2=1.088,0.123 respectively,all P>0.05).(3)When it refers to the G57S locus of Endothelin B receptor gene,the difference of genotype frequencies of GG,GA and AA types between patients with KD and controls were not significant(χ~2=0,P>0.05).There was no statistical significance for the G and A allele frequency between these two groups. And neither the genotype nor allele frequency was observed significant difference between the KD with CAL and without CAL groups(allχ~2=0,P>0.05).Conclusion(1)The polymorphisms of K198N locus of Endothelin-1 gene and the G57S locus of Endothelin B receptor gene had not been found association with the pathogenesis of KD and its complication of coronary artery lesion.(2)The polymorphisms of 70C>G locus of Endothelin A receptor gene might related with the pathogenesis of KD,but there was not significant difference in the polymorphism between KD and that complicated with CAL.
