Background: Breast carcinoma is one of the most frequent malignant tumor. It has been well documented that angiogenesis is one of the most crucial step facilitating tumor growth. Angiogenesis helps to transport oxygen and nutrients to the tumor, which are essential for tumor development. In a number of studies, it has been shown that tumor angiogenesis is correlated with tumor malignancy and patient prognosis. People pay more attention to controning tumor angiogenesis recently. Angiogenesis is tightly regulated by a network of pro-angiogenic and anti-angiogenic. vascular endothelial growth factor is one of the most important pro-angiogenic factor, it can stimulate vascular endothelial cell to caryocinesia, to promote new vessel neogenesis when combinating with VEGFR-2. Meanwhile, it facilitates tumor diffusion in lymphatic system. Vascular endothelial growth inhibitor (VEGI) is a novel anti-angiogenic cytokine which belongs to the tumor necrosis factor (TNF) superfamily and has been identified as a highly effective endogenous inhibitor for endothelial growth and angiogenesis. Very little is known about the significance of VEGI and correlation with VEGF in cancer.Objective: The expression of VEGF and VEGI in breast carcinoma and their correlation with clinicopathological features.Methods: The carcinoma tissues of breast carcinoma were harvested from 32 cases with complete clinic pathological records. The mRNA transcription and protein expression of VEGF and VEGI were detected by RT-PCR and Western blot respectively.Results: Both mRNA transcription and protein expressions of VEGF and VEGI were high relative respectively. There was positively correlative between VEGF and VEGI in tumor tissues. The expression of VEGF was significantly difference according to diameter, metastasis lymph nodes and pathological staging. The expression of VEGF was correlativity in different maximum diameter, metastasis lymph nodes and pathological staging. The expression of VEGI were no differences among different age, ER, PR, C-erb-2, maximum diameter, metastasis lymph nodes and pathological staging, except different pathological types, with lower VEGI in infitrating ductal carcinoma. The expression of VEGI were no correlativity in different age, ER, PR, C-erb-2, maximum diameter, metastasis lymph nodes and pathological staging, But it was correlativity with different pathological types. VEGF/VEGI was no correlativity in different age, ER, PR, C-erb-2. There were positive correlativity between VEGF/VEGI with different pathological types, maximum diameter, metastasis lymph nodes and pathological staging.Conclusion: VEGF has important influence on infiltration and metastasis of breast carcinoma. VEGI is one of important negative regulators controlling angiogenesis of breast carcinoma and acts as a normal physiological role in maintaining a stable vasculature. It is demonstration that not only VEGF but also VEGI are contributed to happen and development of malignant breast tumor. As an endogenous ant-angiogenesis factor, VEGI may use as an important pole in clinic.
