Ilex cornuta, which is evergreen in holly family, distributes in Zhejiang,Jiangsu,Anhui Jiangxi,Hubei,Hunan,Henan,Guangxi, etc. Its tender leaves can be used as tea after it is dunked and insolation, which can cure headache and dispel fever. Its natural sapor is bitter and sweet and it has the function of dispelling heat,cooling blood,nourishing liver and kidney and getting rid of annoyance thirst. There are plenty of reports and research of the active components of Ilex cornuta's leaves. There are many ways of extraction and isolation. Extraction with solvent,distilling with water vapour and so on are included in the traditional way of extraction, while supersonic extraction,microwave extraction and so on are included in modern means of extraction. There are two sorts in isolation and purification generally, of which one is common way of isolation, such as extraction,recrystallization and so on, and the other is chromatography including column chromatography,thin layer chromatography,paper chromatography,gas chromatography and high performance liquid chromatography.But there's no report about its research of Ilex cornuta's roots for the moment. Therefore, the Ilex cornuta's roots were tested as material by our research team to extract,isolate,purify,identify active components in it. And the components' physical-chemical characteristics and chemical structure were studied. The following tasks have been done.1. One kind of organic acid was extracted by extracting with ethanol and then isolated with water. During the extraction process, traditional solvent extraction and supersonic extraction methods were used respectively.Conditions of traditional solvent extraction of the experiments were as follows: extraction temperature 30℃, extraction time 30min;the ratio of ethanol to roots of Ilex cornuta 5:1(v/w). Conditions of supersonic extraction were as follows:extraction temperature 30℃, extraction time 24 hour; the ratio of ethanol to roots of Ilex cornuta 5:1(v/w).2. Two substances were obtained and one of which is white powder and can be dissolved in methanol, ethanol, ethyl ether, chloroform, hot acetic acid and 2% solution of sodium hydroxide in ethanol, and can be easily dissolved in acetone and Pyridine.Its structure described as picture A was primarily identified by IR,NMR,MS.And another component whose structure was described as picture B was ascertained to be Dibutyl phthalate by IR,NMR,MS.To make sure it we synthesized Dibutyl phthalate,whose results were very good. The reported color of Dibutyl phthalate was colorless,but it was blue in this experiment.The reasons for it may be that the impurity effected the color and the state. 3. Roots of Ilex cornuta were treated as raw material in the experiment, which avoided the influence of the plenty of pigments and simplified the process of extraction and isolation to a large extent. What's more, it initiated a new direction for the exploitation of Ilex cornuta and provided us a new way to obtain active components, such as Ursolic Acid.
