Ilex cornuta Lindl. et Paxt., one of the species in the Ilex genus of theAquifoliaceae family. The leaf of this plant, which is recorded as “Ilicis cornutaefolium” in Chinese pharmacopoeia, as well as its root, bark and fruit, can be used asmedicine. Ilicis cornutae folium is the major source of “Ku-Ding-Cha”. The root of thisplant is called “Gong-Lao-Gen” in Chinese medicine, bitter and cool-natured, which caninvigorate the liver and the kidney, expel wind and heat, can cure weakness of waist andknees, arthrodynia, head-wind, acute conjunctivitis, toothache and urticaria. The majorchemical constituents contained in Ilex cornuta are triterpenes, triterpenoid saponins,flavones, flavonoid glycosides, polyphenols and their derivants and fatty acids. Themodern pharmacology research showed that this plant had many efficacies, such ashypolipemic, antibacterium and antifertility. The result of pharmacodynamicsexperiments administered by our team confirmed that the extraction of Ilex cornuta hadthe remarkable bioactivities in antioxidation of myocardial cells, expansion of coronaryartery and hypotensive, especially the n-butanol extraction. There is scarcely any reportof the root of Ilex cornuta about chemical constituents and modern pharmacologicalactions while the researches focus on its leaf.This project was carried out in order to exposit the material base, mainlycontaining separation and purification of ethyl acetate and n-butanol extractions of Ilexcornuta, identification of the achieved compounds.29compounds were separated andidentified, which were2,6-Dimethoxyquinone (GL-1), Urs-12-en-28-oic acid,3-(a-L-arabinopyranosyloxy)-19,23-dihydroxy-,(3,4a,20)-(GL-3), Hexadecanoicacid (GL-5), Gouguside1(GL-7), Chikusetsusaponin IVa butyl ester (GL-10),-D-Glucopyranosiduronic acid,(3β)-17-carboxy-28-norolean-12-en-3-yl2-O-a-L-arabinopyranosyl-,6-butyl ester (GL-11), Lupeol (GL-12), a-Amiradienol(GL-13), a-Amyrine (GL-14), Urs-12-en-28-oic acid,3-(-L-arabinopyranosyloxy)-19-hydroxy-,(3,20)-(GL-15), Oleanolic acid (GL-17),Gousugenin L (GL-19),(Z)-9-Hexadecenoic acid (GL-21), Siaresinolic acid (GL-23),Pomolic acid (GL-24), Gouguside L1(GL-26), Gouguside L (GL-27), Oleanolic acid 3-O-β-D-glucuronopyranoside-6-O-methyl ester (GL-28), Ursolic acid (GL-30),Gouguside L2(GL-32), β-D-Glucopyranosiduronic acid,(3β)-17-carboxy-28-norolean-12-en-3-yl2-O-β-D-arabinopyranosyl-,6-methyl ester(GL-34), β-D-Glucopyranosiduronic acid,(3β)-17-carboxy-28-norolean-12-en-3-yl2-O-a-L-arabinopyranosyl-,6-butyl ester (GL-36), Ursolic acetate (GL-37), Mateside(GL-38), Brevicuspisaponin3(GL-39), Olean-12-en-28-oic acid,3-(α-L-arabinopyranosyloxy)-19-hydroxy-, β-D-glucopyranosyl ester,(3β,19α)-(GL-40), Gouguside7(GL-41), β-Sitosterol (GL-42), β-Sitosteryl glucoside (GL-43).The eight compounds: GL-3, GL-11, GL-15, GL-19, GL-26, GL-27, GL-32,GL-36were new compounds, and the four compounds: GL-19, GL-26, GL-27, GL-32were new skelecton compounds. The seven compounds: GL-1, GL-5, GL-10, GL-13,GL-21, GL-28, GL-34were obtained from this genus for the first time, the sixcompounds: GL-14, GL-23, GL-24, GL-38, GL-39, GL-40were obtained from thisspecies for the first time.In addition, the medicinal resources of Ilex genus and the research of theirchemical constituents and pharmacological actions in the recent10years were reviewedin this paper.This project preliminarily exposited the material base of the root of Ilex cornuta,improved its research of chemical constituents, and provided experimental basis forfurther research and utilization.
