Ilex cornuta Lind, one of the species in the Llex genus, is an evergreen plant Whichdistributes widely in the middle reaches area of Yangtze River provinces. Leaves is one of thesources of Kuding tea, which has the effect of heat clearing, nourishing Yin, reducinghyperactive liver-yang and nourishing the kidney(RLNK), and it can be used for illness suchas hemoptysis induced by pulmonary tuberculosis, steaming of bone, dizziness, andhypertension, and so on. According to the National Chinese herbal medicine records, the rootof the Ilex cornuta has the effect of dispelling wind and relieving pain, and can be used forrheumatism., lumbar muscle strain, headache, toothache, and dice hepatitis. The root is achinese medicinal materials of great medicinal value. There are various of folk prescriptionsin folk, and has obvious therapeutic effect. Currently, research on the chemical constituentsof Ilex cornuta is not enough, in order to known the chemical constituents and get moreactive nature products, we do the research as follows:1. Extraction, separation and purification constituents from the root of Ilex cornuta:extracted the sample with70%ethanol solution repeatedly at room temperature, concentratedthe extract, then floated the extractum in water, extracting with petroleum ether and ethylacetate respectively. We separated and purificated the extracts with several chromatographicmethods, and got five compounds.2. Identification of constituents: compounds were identified by means of determinationof physicochemical constants and spectral data analysis. They were3β-palmitoyl-11-carboyl-urs-12-ene(1)、3β-palmitoyl-7,11-dicarboyl-urs-12-ene(3)、β-stiosterol(4). Compound3is a new triterpene ester compound. The structures of compound2and3still need furtherconfirmed.3. optimization of extraction condition and determination of triterpenoids from the rootof Ilex cornuta: with the absorbance as the reference standard, and selecting ursolic acid asthe reference substance,we investigated the impact of four factors on the extraction oftriterpenoid by ultraviolet spectrophotometry, and the four factors are temperature(A)、 extraction time(B)、solid-liquid ratio (C) and ethanol concentration(D). The result showedthat the primary and secondary factors affecting the absorbance followed by A>B>D>C, andthe optimum conditions were established. When the material was extracted with90%ethanol3h at90℃and at the ratio of liquid to material(g/ml)1:20, the content of triterpenoids fromthe root of Ilex cornuta was3.20%. the research will provide a theoretical basis for thefuture development.
