Ilex cornuta Lindl. et Paxt.(Aquifoliaceae) is mainly distributed in the middle and lower reaches of the Yangtze river region in our country, its leaf has been used as traditional Chinese medicine in China Southern and southwestern for thousands years in folk. Modern pharmacological studies have shown that this plant had many efficacies, such as anti-atherosclerosis, antithrombotic, hypolipemic, and antihypertensive as well as certain effect on cardiovascular and cerebrovascular diseases.In order to further define its relationship between the structure of chemical compounds and pharmacologically active, the separation and purification of the stems of Ilex cornuta, and structure identification work were herein performed. As a result, using various separation techniques, such as silica gel column chromatography, Sephadex LH-20, macroporous resin, hole resin, medium pressure column chromatography, dynamic axial compression chromatography and semi-preparative liquid chromatography, 27 compounds were separated and purified from 60% ethanol extraction of I. Cornuta. In addition, the structure of compounds were identified by 1D- and 2D-NMR and MS spectral analysis. In the twenty seven substances, compounds GM-1、GM-2、GM-20、GM-21、GM-26、GM-31、GM-33 were first isolated from this genus, compounds GM-4、GM-8、GM-19、GM-23 were obtained from this species for the first time, GM-5、GM-7 were new compounds, and their structures were determined as: ursane-12,19-dien-28-oic acid 3β-O-β-D-glucuronopyranoside-6-O-methyl ester, 3α,23α-dihydroxy-olean-9(11),12-diene-28-oic acid 28-O-β-D-glucopyranosyl ester.On the basis of chemical composition study, the isolated compounds GM-1、GM-5、GM-7、GM-12 were tested against H2O2-induced H9c2 cardiomyocyte injury, and the data showed that compounds GM-1、GM-5、GM-7、GM-12 had no significant cell-protective effects.This subject further enrich and perfect the study on the type and quantity of the chemical constituents of the stem of Ilex cornuta, to further explore the phyarmacological effects of Ilex cornuta stems, expect to find new bioactive compounds and elucidate its material basis. We are hoping to develop new drugs for the treatment of cardiovascular and cerebrovascular diseases, providing some material foundation for the later research, and open up a new way for the development of Ilex cornuta.
