Objective: To study the effect of therapeutic alliance of radiofrequency ablation (RFA) and high-frequency thermotherapy(HFT) on the invasion of cony liver cancer cell line VX2. and to evaluate its validity, safety and utility.Methods:32 cony liver cancer VX2 model was established by transplantion, according to the different treatment methods ,the models were divided into four groups, Group A,BB,C,D..each group had 8 cony liver cancer VX2 model. Group A were vacant without any therapeutic intervention. Group B were treatad with therapeutic RFA only at the frist day of experiment. Group C were treated with therapeutic HFH at the frist,fourth,seventh day after the experiment had been started. Group D were treatad with RFA therapeutic alliance of radiofrequency ablation (RFA) and high-frequency thermotherapy(HFT), which were treated with therapeutic RFA only at the frist day of experiment, and were treatad with therapeutic HFH at the frist,fourth,seventh day after the experiment had been started. To evaluate safety of treatment,ALT and AST were detected by drawing out blood at one day before experiment and the frist,fourth,seventh,fourteenth day after the experiment had been started; To evaluate the growth of tumorous, gross tumor volume and tumorous growth rate were calculated by using ultrasound to measure the longest and shortest diameter of tumors one day before experiment and fourteenth day after the experiment was started; recorded the information of metastasis in liver,lung and lymphoid node. Observe the change of pathology by HE dyeing. Evaluat tumors'invading ability to stroma by using immunity class method to detect positive express of matrix metalloprotei-nase(MMP-2,MMP-9) in central tissue of tumor,marginal tissue of tumor and normal hepatic tissue. Evaluat the information of micrangium of tumors by using immunity class method to detect positive express of blood vessel endothelium factor CD34 in central tissue of tumor,marginal tissue of tumor and normal hepatic tissue.Result:1.The level of ALT,AST of each group had no significant difference before treatment(P﹥0.05); The level of ALT,AST of each therapeutic group upgrade significantly, the upgrading amplitude of aminopherase were maximum in group B and group D. The level of ALT,AST of each therapeutic group begin to descend at the fourth day , The level of ALT, AST of group B and group C descended to preoperative level.2.The tumors'growth rate of vacant group was +(28.84±14.13)%,significant super-to every other group(P﹤0.01); The tumors'growth rate of therapeutic alliance of RFA and HFH was -(155.65±52.17)%, which was significant lower compared with solo therapy(P﹤0.05).3.pulmonary metastasis was the most, metastasis rate of RFA+HFH group was 37.5%, RFA group was 62.5% and HFH group was 50.0%, significant difference appeared when compared with control group . metastasis rate of RFA+HFH group compared with that of RFA group has significant difference as well as with that of HFH group.4.Pathology Observation : tumor cellular proliferation, karyome- galy of cellular nucleus,anachromasis were evidently observed through microscope in vacant group,much abnormal division were also observed , hyperplastic collagen fibers were not observed in circum- tissue; Coagulationnecrosis were presented in center of tumor tissue in RFA group as well as in allied group, a great quantity hyperplastic collagen fibers were also observed around centra area , and inflammatory reaction zona is around periph area. Relic tumour cell can be seen by chance in periph area of radiofrequency focus, The area of cellular necrosis and trauma in RFA group was less than that in allied group; coagulation necrosis not observed in tumor tissue of HFH group ,but there was widespread inflammatory reaction in tumor tissue and periph area.5. Positive express rate of MMP-2,MMP-9,CD34: con-gray scale parameter(con-gray scale/ scanning area)In central tissue of tumors, Positive express rate of each index is highest in vacant group and was lowest in groups adopting radiofrequency therapy, Positive express rate of each index in all therapeutic groups compared with that of vacant group was different significantly (P<0.01)In the marginal tissue of tumors , Positive express rate of each index is highest in vacant group, Positive express rate of each index in all therapeutic groups compared with that of vacant group was different significantly (P<0.05), Positive express rate of each index was lowest in allied group, which compared with that of RFA group was different significantly as well as with that of HFH group. (P<0.05).In normal hepatic tissue, Positive express rate of each index is also highest in vacant group, Positive express rate of each index in all therapeutic groups compared with that of vacant group was different significantly (P<0.05), Positive express rate of each index was lowest in allied group, which compared with that of RFA group was different significantly as well as with that of HFH group. (P<0.05). Conclusion: Therapeutic alliance of radiofrequency ablation and highfrequency thermotherapy on cony liver cancer cell line VX2 has only temporary influence on Hepatic function . This experiment adopts merit of radiofrequency ablation, which sacrifice tumours by creating coagulation necrosis under higer megat-emperature, Thus deactivat tumor cells of treatment region thoroughly, Meanwhile, the experiment integrates the merit of high-frequency thermotherapy, which can more stimulates immunity of organism under lower megatemperature . Therapeutic alliance of radiofrequency ablation and high-frequency thermotherapy can suppress expanding of tumor's volume more obviously; The effect of reducing tumorous metastasis is more evidently. And it can more degrade the positive express of MMP-2,MMP-9 and CD34, thus degrade invasiveness of tumors.This is an effective method of treating tumor of liver, It is much more effective than simple therapy and has value of clinical application invariably. And this experiment provides with theoretical base for therapeutic alliance of kinds of thermotherapies.
