Chinese medicinal materials Magnolia officinalis is Magnolia officinalis Rehd.et Wils, or Magnolia officinalis Rehd.et Wils.var.biloba Rehd.et Wils. or Magnolia rostrata W.W.Smith's dry cortex of bole, root and branch. Its mainly effective contents are magnolol and honokiol.This paper is to aim directly at magnoliae cortex of study from dujiangyan in. Sichuan province and HPLC is mainly applicated. Thus we have to feel for the condition of HPLC whose emphasis and difficulty is at the choose of the ratio of mobile phase. Here, I will introduce a part of the condition .HPLC analysis was conducted with a mobile phase containing acetonitrile(B2)-1%Acetic acid (A2) solution and at a flow rate of 1 mL/min. A Diamonsil C18 column (25cm×4.6mm) of 5μm particle size was used for the analysis. Analysis was done at UV 294nm, and 20℃. 0min (10%B2), 0~30min (10%~30%B2), 30~60min (30%~55%B2), 60~70mm(55%~65%B2),70~80mm(65%~85%B2),80~100min(85%~100%B2), 100~110min (100%B2) .The detectable level was 10μL. At last, we got a well HPLC collection of illustrative plates, components can be separated well.Under the condition of optimization, the content of all samples(the extract of magnoliae cortex extracted by methyl alcohol)was determined and analyzed. Using the optimization chromatogram condition, via HPLC research the magnolol and honokiol content of different tree age, part (dry cortex of bole, root, branch and leaf). By the content of magnolol and honokiol dynamic distributing, biological quantity of different tree age, choosing the best collection time and parts, so as to well used Magnolia officinalis Rehd resource.Besides, we found the totally hydroxybenzene content of leaves exceed 2% which the tree age is 25year upwards, reach the quality goods quality criterion of 2005 edition China pharmacopoeia. So the leaves which the tree age is 25year upwards can be used, whenas Magnolia officinalis Rehd leaves are not used previously, wasting a mass of resource. Via HPLC researching the magnolol and honokiol content of leaf, providing academic basis for Magnolia officinalis Rehd leaves qua medicinal materials.
