| Houpo is the dried stem bark,root bark and branch bark of Magnolia officinalis Rehd.et Wils.or Magnolia officinalis Rehd.et Wils.var.Biloba Rehd.et Wils..Listed in the 2015 edition of Chinese Pharmacopoeia,Magnolia officinalis is widely used in clinical treatment for depression,intestinal obstruction,chronic hepatitis,relieve the symptoms of menopause,etc.Magnolia officinalis is of significant medicinal value.Magnolia officinalis is contained in many classic prescriptions such as Dachengqi Decoction,Houpuwenzhong Decoction.Magnolia resources are widely distributed in the southwest of Sichuan,west of Hubei,south of Shaanxi,south of Gansu,etc.Sichuan is one of the main producing areas of Magnolia officinalis,and its productions of Magnolia officinalis are referred to as"Chuanpo",one famous traditional geo-authentic crude drug.Chuanpo resources can be found all over Sichuan Province,such as Dujiangyan city,Leshan city,Aba Prefecture,Mount Emei,Meishan city,Ya’an,etc..As for now there are many reports about the germplasm resources,chemical constituents,pharmacological actions and quality control of Magnolia officinalis,but there is no systematic report on the quality comprehensive evaluation of Magnolia officinalis in Sichuan.For that reason,this study will be carried out in the following aspects:First,to establish HPLC fingerprint of Magnolia officinalis in different regions of Sichuan.42 samples of Magnolia officinalis from different habitats in Sichuan were collected,through optimizing sample preparation methods and studying the chromatography conditions,a method for determination of separation of HPLC fingerprint was established.Based on fingerprint peaks of Magnolia officinalis,identified 9 peaks as common peak for samples.Using the UV spectrum of control of the relative retention time and the on-line collection as the basis,it is confirmed that the HPLC fingerprint of Magnolia officinalis in peak 1,peak 5,peak 8,peak 9,are respectively syringin,magnoflorine,honokiol,magnolol.Through analysis and clustering similarity evaluation of 42 Magnolia sample fingerprint,the similarity was more than 0.8,showing that the difference of chemical composition of Sichuan Magnolia officinalis in different areas is smaller;cluster analysis results showed,except Dujiangyan Long Chi Town Hongkou scenic spot Guanfenggou and Mount Emei scenic gully in Fuxi townFengshou village these two producing area,the rest of the sample clustering is relatively good,and that illustrateed Magnolia officinalis have consistency.Therefore,the scientific method that HPLC fingerprint was developed in this paper,can be used for reasonable consistency of Sichuan Houpo.Second,the identification and analysis of the volatile constituents of Magnolia officinalis in Sichuan:through the use of“content of volatile oil determination”in Chinese Pharmacopoeia(2015 Edition)to determine the content of volatile oil in Magnolia and the use of GC-MS method to identify 42 areas of volatile components.From the volatile oil yield analysis,magnolia produced in Meishan city Hongya County Zhang village and Mount Emei City,Long Chi town honghua village,the oil content was higher,with the content of 0.4%;however,the magnolia produced in Mount Emei City Fu Xi town Fengshou village and Leshan City,Jiajiang county Huang Ping Village,the content of volatile oil is low,only 0.17%.Therefore,the results show that the volatile oil content of Magnolia officinalis from different regions of Sichuan varied greatly.From the analysis of volatile components in different regions of Sichuan,153 chemical constituents were determined among 42 Magnolia officinalis volatile oil samples,the main composition as terpene,terpene alcohol,sesquiterpenoids,Higher aliphatic alkane,oxy-derivatives,etc.5 compounds were the common components for Magnolia from different producing area,respectively caryophyllene,1-alpha caryophyllene,alpha muurolene.12-Oxabicyclo[9.1.0]dodeca-3,7-diene,1,5,5,8-tetramethyl-,(1R,3E,7E,11R,2-Naphthalenemethanol,1,2),3,4,4a,5,6,7-octahydro-a,a,4a,8-tetramethyl-,(2R,4aR)-.The results showed that there were significant differences in the content and composition of chemical components in Magnolia officinalis from different habitats in Sichuan.Third,the contents of active ingredients in Magnolia officinalis were determined by HPLC,acid-base titration and spectrophotometry.The contents of phenols,alkaloids and polysaccharides in 42 batches of Magnolia officinalis were determined.The contents of magnolol was 8.5636.41 mg·g-1,and the content of honokiol was5.2929.17 mg·g-1.The total amount of magnolol and honokiol was 14.3665.58mg·g-1,which was consistent with the determination of magnolol and honokiol in pharmacopoeia.The contents of syringin and magnoflorine were respectively 0.01%0.22%,0.01%0.21%;the total alkaloid content was 2.71986.9111 mg·g-11 and the content of polysaccharide was 3.89%21.51%.The difference of chemical composition in Magnolia officinalis from different areas of Sichuan is more significant.Fourthly,the comprehensive evaluation of the chemical quality of Magnolia officinalis in Sichuan was established:Grey relational analysis.By using gray correlation analysis method to study the effects of honokiol,honokiol,volatile oil,total alkaloid,polysaccharide,the quality of Magnolia officinalis and magnolol,and the total amount of magnolol as evaluation index were used to evaluate the chemical quality of Magnolia officinalis in Sichuan.Results showed the relative correlation of42 batches of medicinal herbs was between 0.282 and 0.597,which indicated that the chemical quality of Magnolia officinalis was different.Finally,the research progress of chemical constituents and germplasm resources of Magnolia officinalis was reviewed.The results of this study provide a scientific basis for the quality comprehensive evaluation of Magnolia officinalis,the breeding of improved varieties and the protection and utilization of medicinal resources. |
