ObjectiveTo observe the effects of the cluster needling of scalp acupuncture on the neurological function score and the expression of myelin basic protein(MBP)at different time points(24h,3d,7d)in rats with acute focal cerebral ischemia,and to investigate the possible mechanism of the cluster needling of scalp acupuncture on the protection of nerve function in rats with acute cerebral ischemia.MethodsSeventy-two male SPF-grade SD rats were randomly divided into four groups,named false operation group(A,n=18),model group(B,n=18),acupuncture group(C,n=18),acupuncture pretreatment group(D,n=18).Every group was divided into three groups(6 in each group)accorrding to time points of 24h?3d?7d.The local cerebral ischemia rats model in Group B,C and D were prepared by modified suture-occluded method,while Group A was sham model.Rats in Group C were treated with the cluster needling of scalp acupuncture at different time points of 24h,3d and 7d after MCAO.Rats in Group D were treated with the cluster needling of scalp acupuncture for 7 days before MCAO.Group A and B did not receive treatment intervention.Bederson neurological function scoring method was used to score the rats in different time poins 4h?24h?3d and 7d.The expression of myelin basic protein(MBP)in hippocampal tissues of rats in each group was detected by Western-blot.Results1.Neurological function scoreInter-group comparison:In the point of 4h,the neurological function score in Group B,C and D compared with that in Group A was statistically significant different(P<0.05).There was no significant difference between the acupuncture group(Group C)and the model group(Group B)(P>0.05).Compared with the model group(Group B),the neurological function score of the acupuncture pretreatment group(Group D)decreased(P<0.05).In the point of 24h,compared with the model group(Group B),there was no significant difference in the neurological function score of the acupuncture group(Group C)(P>0.05),and the neurological function score of the acupuncture pretreatment group(Group D)decreased(P<0.05).In the point of 3d,compared with the model group(Group B),there was no significant difference in the neurological function score in the acupuncture group(Group C)and the acupuncture pretreatment group(Group D)(P>0.05).In the point of 7d,compared with the model group(Group B),the neurological function score of the acupuncture group(Group C)decreased(P<0.05),while the neurological function score of the acupuncture pretreatment group(Group D)had no significant change(P>0.05).Compared with the false operation group(Group A),the model group(Group B),the acupuncture group(Group C)and the acupuncture pretreatment group(Group D)showed statistically significant differences in 24 h,3 d and 7 d(P<0.05).Intra-group comparison:After observing the three time points of 24 h,3 d and 7 d,it was found that the neurological function score of acupuncture group(Group C)decreased with the increase of treatment time,but the difference was not statistically significant(P>0.05).There was no significant difference in neurological function score of model group(Group B)at 24 h,3 d and 7 d(P>0.05).Also,there was no significant difference in neurological function score of acupuncture pretreatment group(Group D)at 24 h,3 d and 7 d(P>0.05).2.The expression of MBP tested by Western-blotInter-group comparison:In the point of 24h,compared with false operation group(Group A),MBP expression in model group(Group B)and acupuncture group(Group C)increased(P<0.05),but there was no significant change in acupuncture pretreatment group(Group D)(P>0.05).Compared with the model group(Group B),there was no significant change in MBP expression in the acupuncture group(Group C)(P>0.05),while the MBP expression in the acupuncture pretreatment group(Group D)decreased(P<0.05).In the point of 3d,compared with false operation group(Group A),MBP expression in model group(Group B)and acupuncture group(Group C)increased significantly(P<0.05),but MBP expression in acupuncture pretreatment group(Group D)did not change significantly(P>0.05).Compared with the model group(Group B),there was no significant difference in MBP expression in the acupuncture group(Group C)and the acupuncture pretreatment group(Group D)(P>0.05).In the point of 7d,compared with false operation group(Group A),MBP expression in model group(Group B)increased significantly(P<0.05),while MBP expression in acupuncture group(Group C)and acupuncture pretreatment group(Group D)did not change significantly(P>0.05).Compared with the model group(Group B),the expression of MBP in the acupuncture group(Group C)decreased(P<0.05),but there was no significant change in the expression of MBP in the acupuncture pretreatment group(Group D)(P>0.05).Intra-group comparison:Observation of 24 h,3 d and 7 d showed that MBP expression in acupuncture group(Group C)decreased with the increase of treatment time,but the difference was not statistically significant(P>0.05).There was no significant difference in the expression levels of false operation group(Group A),model group(Group B)and acupuncture pretreatment group(Group D)in 24 h,3 d and 7 d(P>0.05).Conclusion1.The method of cluster needling of scalp acupuncture can reduce the score of nerve function and promote the recovery of nerve function in MCAO rats.The pretreatment of the cluster needling of scalp acupuncture before rat modeling can significantly improve the nerve function in the early stage and the therapeutic effect gradually weakened with the passage of time.After rat modeling,the effect of the cluster needling of scalp acupuncture intervention gradually increases with time.2.After MCAO modeling,the expression of MBP increased significantly in rats,and the expression of MBP could be down-regulated by the intervention of the cluster needling of scalp acupuncture,suggesting that the cluster needling of scalp acupuncture may stabilize the structure of myelin sheath by down-regulating the expression of MBP,thus playing a protective role on brain tissue.And the pretreatment of the cluster needling of scalp acupuncture in rats before model establishment can play a therapeutic role in the early stage.After model establishment,the intervention of the cluster needling of scalp acupuncture in rats needs a certain time to accumulate and play a therapeutic role.
