Oral Lichen Planus(OLP)is the common disease of mouth cavity diseases, which brings patients tremendous pain.Current research shows that the function mess of the cell immunity is the characteristic pathological change,but the mechanism of its immunity mess still unknow,At present the role of CD 4+ T cells and Th1 cell factors in the immune diseases are the hot research spots.Osteopontin,(OPN)is a Th1 cell factor,which is discovered at latest.it can be produced by various organizations and cells then makes many effects.OPN is a prominent matricellular component of extracellular matrix.Combined with integrin receptors(ανβ3)or CD44 through RGD pathway and non-RGD pathway(do not depend on RGD structure),OPN mediates cell adhesion and migration etc.various cell behavior,it also plays an importantly role in the cell immunity,wound heal,the tumor metaptosis,OPN is an important mediate factor in the process of development of body and organs.It has reported recently that OPN participates in inflammation and immunie responds,also plays an important role in the generation and development of rheumatism arthritis and autoimmunity diseases.But it has not be reported in oral mucosa diseases.We exam serum OPN protein concentration and the expression of CD44 of eripheral blood CD4+T lymphocytic subpopulations of OLP,and patients' serum IL-8,IL-12 proteins concentration,We discuss the role of OPN and and its receptor CD44 in the pathogenesis and progress of OLP.Objective: (1)To determine the expression of OPN on the local mucosal epithelium and CD44 of eripheral blood CD4+T lymphocytic subpopulations of OLP patients.(2)To exam the serum concentration of OPN,IL-8,IL-12 of OLP,and analyze the relativity with pathogenesis of OLP,it can reveal OLP mechanisms and provide an experiment basis for building up a new treatment method of OLP further.Methods:45 OLP patients and 20 healthy volunteers coming from Shandong University stomotology hospital(1)Detect local mucosal epithelium OPN expression through immunity histochemistry strain.(2)Exam serum OPN concentration through ELASA method;(3)Detect CD44 of eripheral blood CD4+T lymphocytic subpopulations through flow cytometry;(4)Investigate sereum IL-8,IL-12 concentration through liquid chip;T test was used for statistical analysis.Results:(1)The expression of OPN was expressed on oral mucosal epithelium of OLP patients,respectively.(2)OLP patients had higher levels of OPN concentration than that of control group. (P<0.05).(3)To detect CD44 of eripheral blood CD4+T lymphocytic subpopulations:OLP patients had higher levels of CD44 than that of control group.(P<0.05).(4)To observe the serum IL-8 and IL-12 concentration:OLP patients had higher levels of IL-8 and IL-12 concentration than that of control group.(P<0.05).Conclusion:(1)This research discovers that the expression of OPN was expressed on oral mucosal epithelium of OLP patients and OLP patients had higher levels of OPN concentration than that of control group,provide that OPN indiced CD4+T cell activition abnormal through CD44 pathway.(2)This research discovers the expression of CD44 of eripheral blood CD4+T lymphocytic subpopulations of OLP patients is prominently higher than control group,presume that CD44 maybe participate in early cell immune of OLP and cell immune response abnormal of inflammation subsequently.(3)This research still discovers that OLP patients has higher levels of IL-8 and IL-12 concentration than that of control group,provides IL-8 plays an important oral in local leukomonocyte infiltrating,Th1 response is in the ascendant of OLP patients,OPN is likely to be key factor.
