Objective:To investigate the effect of Mifepristone(MIF)on cadherin and matrix metalloproteinase expression through nuclear factor kappa B in endometrial cancer.Methods:Endometrial carcinoma cell line HHUA was cultured with different concentrations(0,0.1×10-6,0.1×10-5,1×10-5,5×10-5,1×10-4mol/L)of MIF in vitro. MTT was used to examine the cell growth.Immunohistochemistry assays(SP),Western blot and RT-PCR were used to detect the expression of NF-κBp65,E-cadherin,MMP-2 and MMP-9.Results:MIF inhibited the growth of HHUA cell.The expression of NF-κBp65,MMP-2 and MMP-9 significantly decreased(p<0.05),and the expression of E-cadherin increased obviously(p<0.05).The expression of NF-κBp65 was obviously in positive correlation with the expression of MMP-2 and MMP-9(r12 =0.65,r22=0.89),and it significantly negative correlated with the expression of E-cadherin(r32=-0.94). Conclusions:MIF probably inhibited the invasion of human endometrial cancer cells,and the up-regulation of E-cadherin activity,the down-regulation of NF-κBp65,MMP-2 and MMP-9 activity may account for this invasion.
