The Relationship Between Cycloxygenase-2 And Survivin Expression And Vascularization In Carcinoma Of Large Intestine

Objective:To investigate the relationship between COX-2 and survivin expression and tumor vascularization in cancer of colon,and study on their roles in development of cancer of colon,new vessels formation.To provide valuable biological detecting index for early diagnosis and prognosis in cancer of colon.Method:Streptavidin-peroxidase-biotin immunohistochemistry technique was used to detect the expression of COX-2,Survivin and MVD(labled by CD34) in forty-eight cancer of colon specimens, twenty adenomatous polyp specimens, twenty normal mucosa specimens.The relationship among the expression of COX-2,Survivin,MVD and clinicopathological features in cancer of colon were analyzed statistically.Results: (1)The positive expression rate of COX-2 in cancer of colon (79.17%) was signifficantly higher than in adenomatous polyp(20.00%)and in normal mucosa(10.00%) (P<0.01, P<0.01) ; The positive expression rate of COX-2 in cancer of colon was signifficantly correlated with Dukes stages(P <0. 0l),but incorrelated with sex,age,tumor location,tumor size and degree of tissue differentiation(P>0.05). (2) The positive expression rate of Survivin in cancer of colon (68.75%) was signifficantly higher than in adenomatous polyp(25.00%)and in normal mucosa(15.00%) (P<0.01, P<0.01); The positive expression rate of Survivin in cancer of colon was signifficantly correlated with Dukes stages(P <0. 0l),but incorrelated with sex,age,tumor location,tumor size and degree of tissue differentiation(P>0.05).(3)There was positive correlation between the expression of COX-2 and Survivin in cancer of colon.(4)The MVD value in COX-2 positive group(34. 00±7. 34) was higher than that in COX-2 negative group(23. 96±4. 23),and the MVD value in Survivin positive group(35. 79±6.25) was higher than that in Survivin negative group(23. 60±3. 72),and there was a significant difference(P <0. 0l, P <0. 01); The MVD value in both positive group(36. 98±5. 48) of COX-2 and Survivin was higher than that in both negative group(22. 39±3. 84) of them, and there was a significant difference(P<0.01).Conclusion:1.There was specific expression up-regulation of COX-2 and Survivin in tissue of cancer of colon,and there was positive correlation between their expression and cancer of colon,which suggested that may participated in development of cancer of colon.2.The high expression of COX-2,Survivin in cancer of colon tissue closely related to Dukes stages ,which suggested that they may highly related with prognosis of cancer of colon.3.There was correlation between expression of COX-2,Survivin and MVD;the MVD value in both positive group of COX-2 and Survivin was obviously higher than that in both negative group of COX-2 and Survivin.All of them suggested that they promoted the formation of blood vessel together in cancer of colon.
