Objective: By detection of CK19 mRNA in peripheral blood, we aimed to study the correlation between micrometastasis of breast cancers and tumor size,node metastasis,clinical stage,age, menopause status and expression of receptors. By comparing the variation of CK19 mRNA before and after adjuvant chemotherapy, we aimed to evaluate the effect of adjuvant chemotherapy on controlling the micrometastasis of breast cancers.Method: 60 postoperative patients with breast cancers were enrolled in this study, who were divided into three groups (0~Ⅰ,ⅡandⅢ). CAF scheme were applied on all these patients. CK19 mRNA was detected as marker of peripheral micrometastasis by means of RT-PCR before and after adjuvant chemotherapy individually. Ten patients with benign breast tumors served as negative controls. The data were analyzed by means of chi-square test ,SAS v9.0 software.Result: Every RNA has oneβ-actin strip,and one 196bp strip were corresponding to CK19. CK19 expression was negtive in control group. Total positive rate of CK19 expression in the postoperative patients with breast cancer was 23.33%(14/60) before adjuvant chemotherapy. Post-chemotherapy positive rate of CK19 expression was 3.33%(2/60),which was significantly lower than that of pre-chemotherapy ( 23.33%) (14/60).Positive rate of CK19 expression in three groups varied significantly before chemotherapy individually(p<0.01). Patients with bigger tumors or those who had node metastasis had higher positive rate of CK19 expression in peripheral blood.Conclusion: The positive rate of CK19 expression in the peripheral blood of breast cancer patients had statistically correlation with tumor size,node metastasis and clinical stage,but had few correlation with age,menopause status and expression of receptors. Adjuvant chemotherapy could control micrometastasis of breast cancers and might play an important role on the prognosis of patients.
