Objective: Isolated from primary liver cancer (PLC) and gastric cancer tissues, the Helicobacter were identified to be Helicobacter pylori(Hp). Then their susceptibility and antibiotic resistance were analyzed. The study Provides the valuable reference for the clinical preventing and controlling infection.Method: 1,We selected 28 PLC patients' liver tissues and 25 gastric cancer patients' gastric tissues for the experimental group, 22 other liver disease patients' liver tissues and 36 other gastric disease patients' gastric tissues for the comparison group. We used the Colombian culture medium to isolate and cultivate the Helicobacter in the tissues, then performed appraisal by dyeing, the electricity mirror observation and urease test on the masculine specimen. We carried on the polymerase chain reaction (PCR) to examine Helicobacter 16SrRNA gene as the preliminary identification. 2,Confirmation: the 16SrRNA expanding-production of the tissues and bacteria were performed Southern hybrid and compared by the sequence homology . 3,The susceptibility and antibiotic resistance were tested by E-test and broth agar medium method.Results: 1,21 Helicobacter were isolated in all: the liver cancer tissues 3, the separation masculine gender rate 10.7%; the gastric cancer tissues 6, the separation masculine gender rate 24%; other gastric disease tissues 12, the separation masculine gender rate 77.3%. 2,In 28 PLC specimen 17 cases were extracted Helicobacter 16SrRNA gene, the mas -culine gender rate was 60.7%; in 25 gastric cancers specimen 18 cases were extracted, the masculine gender rate was 72%; in 36 Other gastric disease specimens 28 cases were extracted, the masculine gender rate was 77.8%. It was demonstrated by Statistics that there was no remarkable difference (Hp > 0.05) on the masculine gender rate between the liver cancer group and the gastric cancer group. so was the gastric cancer group and the other gastric disease group (Hp > 0.05); but the difference between the liver cancer group and the other liver disease group was remarkable (Hp < 0.01). Performed by the Gram staining, the electricity mirror, urease test, the 16SrRNA gene examination and the sequence determination , the 21 isolates were confirmed as Hp. The sequence determination indicated that the isolates' Helicobacter 16SrRNA sequence and Hp have 97.80% homology. 3,The Resistance of the 21 Hp (E-test) to metronidazole is 61.9%, amo- xicillin is 19.0%, tetracycline is 0 ,clarithromycin is 4.76%. The result provides the reference for the treatment of the clinical Hp infection.Conclusion: 1,The resistance of Hp (E-test) that isolated from the liver cancer and the gastric cancer in Qingdao area is 61.9% to metronidazole, which is the highest; amoxicillin is 19.0%, tetracycline is 0 ,clarithromycin is 4.76%.This indicates that the tetracyclin is the most sensitive to Hp, the metronidazole is the most insensitive. 2,Basically tally to the sensitive experiment result of the antibiotic medicine commonly used on Hp and the results of other domestic laboratories. The result of, both the 16SrRNA gene examination and the sequence determination , prove that the infections are Hp infections. 3,There are Hp infections in the tissues of PLC and the gastric cancer, and the infection rate is high, which prompts that the infections and PLC and the gastric cancer possibly have certain relevances.
