Objective To evaluate the value of the new tumor marker AMACR combined with CK34βE12 and p63 immunotaining for the diagnosis and differential diagnosis of prostate cancer;to have an understanding of the correlation between the expression of AMACR and the classification of prostate cancer and the expression of PSA;to have a preliminary discussion of the mechanism of AMACR in the occurrence of prostate cancer.Methods Immunohistochemistry was used to detect the expression of AMACR,34βE12,p63 and PSA in prostate cancer(PCa),paraneoplastic normal tissue,benign prostate hyperplasia(BPH),HGPIN and AAH.Then we statistics the positive expression rate of AMACR,34βE12 and p63 in above-mentioned tissue,analyse the value of AMACR in the diagnosis of prostate cancer and the relationships between the expression of AMACR and the classification of prostate cancer and the expression of PSA in prostate tissue.Results The positive expression rate of AMACR in prostate cancer(PCa),paraneoplastic normal tissue,benign prostate hyperplasia(BPH),HGPIN and AAH is 92.7%(38/41),0%,0%,40%(6/15),11.1%(1/9);The positive expression rate of AMACR among the five groups is marked statisticlly significant,P<0.01,compared with PCa group the expression of AMACR in HGPIN group is marked statisticlly significant both in the positive expression rate and intensity,P<0.01.Staining in basal cells show tissue of prostate cancer lack of basal cells,paraneoplastic normal tissue and benign prostate hyperplasia(BPH) has continuous basal cells in most of the glands;Tissue in HGPIN and AAH has continuous or imtermittent basal cells.The intensity of AMACR in prostate cancer has no realationship with the classification of prostate cancer and the expression of PSA in prostate tissue.Conclusion AMACR,which can raise the accuracy of diagnosis in prostate cancer combined with 34βE12 and p63 can be used as a prostate cancer specific marker, especially in the diagnosis of punctured small sample.The detection of AMACR can provid clues and experimental evidence for the proceed,development and the research of treatment for prostate cancer.
