[Objective] 1.To detect the expression of AMACR, p63 and 34βE12 in the same prostate needle biopsy specimens using the immunohistochemical analysis with a triple-antibody cocktail staining and double-color chromogens; 2.Analyze the relationship of the expression between AMACR(P504S) and the histopathological grade, clinical stage of the prostate cancer; 3.To investigate the value of detection of AMACR(P504S), p63, 34βE12 cocktail in the early diagnosis of prostate cancer.[Methods] The expression of AMACR, p63 and 34βE12 in 42 cases of prostate cancer, 12 cases of high-grade prostatic intraepithelial neoplasia (HGPIN) and 30 cases of benign prostatic hyperplasia (BPH) needle biopsy specimens were examined using MaxvisionTM single-step immunohistochemical method with a triple-antibody cocktail (AMACR/p63/34βE12) staining and double-color chromogens in single paraffin sections.The expression of AMACR, p63 and 34βE12 in prostate needle biopsy specimens and the relationship between the intensity of AMACR(P504S) and the prostate cancer's Gleason grading were analyzed with SPSS 11.5 softward.[Results] The expression of AMACR, p63 and 34βE12 were significantly different between the prostate adenocarcinoma and high-grade prostatic intraepithelial neoplasias or benign prostatic hyperplasia (P<0.05).The staining of prostatic adenocarcinoma were negative for basal cell markers and positive for AMACR, the strongly positive rate of AMACR was 95.2%, only 2 cases were weakly positive expression, the level of expression of AMACR was not correlated with prostate cancer's Gleason grading; HGPIN had a interrupted or continuous basal cell layer and weakly positive or positive expression for AMACR; BPH was strong expression for p63 and 34βE12, but negative for AMACR.[Conclusion] l.To detect the expression of AMACR, p63 and 34βE12 in the same prostate needle biopsy specimens using the immunohistochemical analysis with a...
