Objective To study the proliferation of inhibition by Kanglaite in the hepatocellular carcinoma cell line HepG2 in vitro and find its mechanism.To provide the theory foundation that if it can be used for the chemotherapy of hepatocellular carcinoma.Methods The HepG2 cells were cultured in vitro and treated with KLT at different doses(5μl/ml,10μl/ml,15μl/ml,20μl/ml).MTT was used to observe the inhibition effect of KLT on cells growth.Flow cytometry(FCM) was used to detect apoptosis and the apoptosis related change of protein Fas and Fas-L.Real-time fluorescent quantitative RT-PCR method was applied to investigate the quantitative expression of Fas and Fas-L mRNA.Results(1) KLT produced an obvious time- and - dose- dependent inhibition effect on the HepG2 cells.The live cells decreased gradually with the increasing of drug concentration and disposal time.(2) Markedly apoptosis was detected by FCM in HepG2 cells after treatment by KLT;compared with the control group,the apoptosis rate of experimental group had a great increase.(3) The protein of Fas increased 11.01%, 18.71%,28.71%,37.15%,and the protein of Fas-L increased 1.49%,1.91%,3.27%,3.38% in comparison with the control(P<0.05).(4) Real-time fluorescent quantitative RT-PCR showed that the expression of Fas mRNAand Fas-L mRNA was apparently increased on the treating cells with KLT.Conclusion(1)KLT produced an obvious inhibition effect on HepG2 cells and induced HepG2 apoptosis;(2) KLT can increase the expression of Fas and Fas-L protein and the expression of Fas mRNA and Fas-L mRNA.This may probably play an important function in the inhibition and apoptosis of HepG2 induced by KLT.
