| Objective To investigate the expression of osteopontin (OPN) and cell-surface adhesive molecules CD44v6 in invasive cervical carcinoma, and the relationship between the expression and clinicopathological characteristics of invasive cervical carcinoma. To analysis the correlation of OPN and CD44v6, and elucidate their role in tumor invasion and metastasis.Methods 30 cases of CIN(12 cases of CINâ… and 18 cases of CINⅡ~Ⅲ)and 50 cases of invasive cervical cancer were collected in tongji hospital during January 2003 to October 2005, which were received after hysterectomy and treated with no radiotherapy and chem- otherapy before surgery. Besides, 10 cases chronic cervicitis were collected for control group. Immunohistochemical S-P method was performed to detect the expression of OPN and CD44v6 in all of the tissues above. Statistical analysis was conducted using the package SPSS I 1.5 for Windows. Pearson's char-square and matched data correlation analysis were used to analyze the data.Results 1. OPN was mainly located in cytoplasm, and CD44v6 was mainly located in membrane. Positive staining were brown and yellow particles.2. OPN expression rates in chronic cervicitis, CINâ… , CINⅡ~Ⅲand invasive cervical carcinoma were 10.00% (1/10), 25.00%, 44.44%, 60.00%, respectively. CD44v6 expression rates were 10.00% (1/10), 25.00%, 55.55%, 68.00%, respectively. That is, OPN and CD44v6 expression rates increased gradually in the order of chronic cervicitis, CINâ… , CINⅡ~Ⅲand invasive cervical carcinoma. Comparing with chronic cervicitis and CINâ… , OPN expression in invasive cervical carcinoma had significant different(P<0.05) . Comparing with chronic cervicitis and CINâ… , CD44v6 expression in invasive cervical carcinoma had significant differences, too(P<0.05) . Further more, the difference of CD44v6 expression between chronic cervicitis and CINⅡ~Ⅲwas statistically significant. 3. Between cases with and without lymph node metastases a statistically distinct difference in OPN expression was detected, and so was that in CD44v6 expression(P<0.05). Additionally, there were no significant differences in OPN expression and CD44v6 expression in terms of patients'age, histological grading and histological types. From clinic stageâ… toâ…¢/â…£, expression of OPN increased gradually, compared with clinic stageâ… orâ…¡, expression o f OPN was significantly different in stageâ…¢/â…£. From clinic stageâ… toâ…¢/â…£, expression of CD44v6 increased gradually, too. But there was no significant difference between different clinic stages.4. There was a closely positive correlation between OPN and CD44v6 in invasive cervical cancer.Conclusions OPN and CD44v6 may be both involved in the genesis, development, and metastasis of cervical carcinoma. There was a positive correlation between them. Its expression may be important in evaluating of the tumor progression, metstatic status and judging prognosis of the cervical cancer. |
