Cardiovascular disease, known as"the leading killer"in the world, threatened human health seriously , and ischemic heart disease (IHD)could deteriorate into malignant pathological changes ,such as heart disorder and congestive heart failure, so the prevention and treatment of IHD were focused on by current medical circles. At present, the toxic and side effect of chemical synthesis medicine became visible day by day, and the medicine made from natural botanical drugs were emphasized gradually. Many kinds of bioactivators were contained in Hawthorn Leaves, of which flavonoids was effective constituent to cure cardiovascular disease. Our country was rich in hawthorn, and hawthorn leaves was a kind of renewable resource, if we had make the most of hawthorn leaves to research&design and produce new drugs to cure cardiovascular disease, enormous economic and social efficiency would be brought about, so this idea had broad prospects.Methods In this experiment, myocardial ischemic reperfusion injury models were established by occluding the left anterior descending branch of coronary artery for 40 min and removing the ligation later to reperfuse for 120 min in vivo. Different dosages of hawthorn leaves flavonoids (HLF) (7.5, 15.0, 30.0 mg/kg) were administered by i.p.30 min before the onset of ischemia. At the end of each experiment, 120 minutes after administration, electrocardiogram (ECG) was observed continuously and ST segments were measured, the enzymology in serum were determined, including creatine phos-phokinase( CK ) ,Lactatede hydrogenase ( LDH ) , aspartate transaminate( AST ) ,erythrocuprein(SOD) , malonaldhyede (MDA) and nitrogen monoxidum(NO). In addition, the changes of myocardial ischemia size was determined, the effects on cardiac muscle tissue pathological morphology of rats were observed, and the protective effects and mechanism of HLF on infarction myocardium were explored initially.Results HLF could improve ECG of myocardial ischemia reperfusion rats , inhibit the rise of LDH, CK, AST , MDA and increase SOD and NO active content in serum of myocardial ischemia reperfusion rats. In addition, HLF could decrease myocardial ischemia area and pathological lesion of heart. These all showed that HLF could protect myocardial from ischemia.Conclusions HLF could decrease the cardiac muscle tissue injury, improve SOD activity , reduce MDA content in serum on acute myocardial infarction in rats , relieve the injure of free radical to myocardium cell membranes and maintain myocardial membrane structure, inhibit AST, LDH, CK release and reduce the injury to myocardium issues and cells. So the results suggest that HLF had obvious therapeutic effect on acute myocardial infarction and reperfusion (AMI). The mechanisms of its anti myocardial damage after AMI were related to improving the anti free radical ability of organism and anti-lipid peroxidative reaction, and decreasing the cell death because of generation of free radical.The results showed that HLF had the protective effect on myocardial cell in the condition of ischemia .
