Detect expression of MAP2 of SWO-38 cell line in treated group with CDA-2 for 72h and untreated group by immunofluorescence method. Detect expression of MAP2 and PTEN before and after treated with CDA-2 by RT-PCR and Western blot method.Results: 1. MAP2 was expressed differently in nearly all issues of human glioma. And there was negative correlation between the expression of MAP2 and grading of glioma. 2. MAP2 was up-regulated after treated with CDA-2, which was located in cytoplasm and process in cells. And SWO-38 cells treated with CDA-2 had more cytoplasm and longer processes, compared with untreated cells. 3. PTEN was up-regulated in 24h and then decreased following the treatment of CDA-2.Conclusion: 1. Higher expression of MAP2 in human glioma was associated with lower grade and MAP2 could become a useful index in differential diagnosis and grading of glioma. 2. CDA-2 could induce the differentiation of SWO-38 and up-regulate MAP2. 3. MAP2 could be a useful marker for judging the degree of differentiation in glioma and involved in differentiation of glioma. 4. PTEN could regulate MAP2 in SWO-38 cells treated with CDA-2.
