Glioma is one kind of the malignant tumors and also one of the most difficultdiseases to treat too The clinical therapy includes surgery radiotherapy andchemotherapy However the prognosis is still not as ideal as we thought In ourresearch we observed the expression of PTEN pAKT AKT GSK3pGSK3in low grade gliomas high grade gliomas and normal brain tissues by westernblot in order to investigated the relationship of PTEN PI3K/AKT pathway inGliomaMethod Ten glioma tissues were provided from the Wuxi Second Hospitalbetween Feburay and October in2010in our study Four of them were low grade ofglioma (WHO I II) and six of them were high grade of glioma(WHO IIIIV) The control group was normal human brain tissues which gained by patientswho have operation of craniocerebral injury We observed the expression ofPTEN pAKT AKT GSK3pGSK3in low grade glioma high gradeglioma and normal brain tissues by western blot The SPSS180was used toanalyze the relationship of PTEN AKT pAKT GSK3pGSK3in normalbrain tissues and different grades of gliomaResults The western blot showed that1PTEN we can descried thatPTEN had the highest expression in normal brain tissue lowest expression inhigh grade glioma And the low grade glioma was between those two Thedifference of the expression and the gradient were remarkable in those three kinds ofSpecimens(P<005)2pAKT and pGSK3They were highest in high gradeglioma while lowest in normal brain tissues and the low grade glioma was betweenthose two The difference of the expression and the gradient were remarkable inthose three kinds of Specimens(P<005)3The expression of AKT and GSK3 were basically the same in those three kinds of specimensConclusion Based on the results of our study PTEN pAKT and pGSK3have significant difference and obvious gradient in different grades of glioma andnormal brain tissues The expression level of PTEN is negative correlation to themalignancy of human glioma The result also shows a high positive correlationbetween pAKT pGSK3and the malignancy of human glioma1The decrease of the expression level of PTEN may involved in theoccurrence and development of glioma the less of the expression the moremalignancy of human glioma2PTEN exerts its carcinostasis through the PI3K/AKT cell signaltransmission pathway which can confront the activity transforming of AKTindirectly The high expression of pAKT is another indispensable factor in theoccurrence and development of glioma The increase of the expression of pAKT isone of the reasons of the vicious transformation of glioma3GSK3is one of the downstream proteins of PI3K/AKT cell signaltransmission pathway The increase of the pGSK3is also one important reason inthe occurrence and development of glioma the increase of the expression ofpGSK3is another reason of the vicious transformation of glioma4AKT is one of the key proteins in PI3K/AKT cell signal transmissionpathway it doesn’t has biologic activity AKT is not one of the reasons of theoccurrence and development of glioma It is not participate the vicioustransformation of glioma either5PTEN have opposite actions of pAKT and pGSK3The occurrence anddevelopment of glioma is the result of the decrease of PTEN and the increase ofpAKT pGSK3So is the vicious transformation of glioma6The results of this experiment prompt that the occurrence and development of glioma can be controlled if the expression of PTEN pAKT pGSK3beencommanded The treatment effect can be enhanced too...
