Objectives:Through the PTEN protein expression and methylation status of the different levels of PTEN in glioma cells, to explore the relationship between PTEN gene methylation and glioma, and provide support for the early diagnosis, prognosis judgment and gene therapy of glioma.Methods:24cases which proved to be glioma by pathology came from Third Xiangya Hospital and Hunan Provincial of Tumor Hospital. Patients were first collected specimens of the disease, and have not received chemotherapy before surgery, radiotherapy and immunotherapy before surgery. There are a complete clinical data. there were eight cases of grade Ⅰ-Ⅱ, four cases of grade Ⅱ, four cases of grade III and eight cases of grade Ⅳ. Classified Ⅰ-Ⅱ grade gliomas were classified as low-grade gliomas and Ⅲ-Ⅳ grade gliomas were classified as high-grade gliomas.8cases of non-tumor and normal brain tissue were taken from the Third Xiangya Hospital. First,PTEN protein expression was detected in glioma tissues and normal brain tissue.Second, methylation status of PTEN gene was detected in glioma tissues and normal brain tissue.Results:1.the difference of PTEN protein expression was significant between normal brain and low-grade gliomas (P<0.05). the difference of PTEN protein expression was significant between normal brain and high-grade gliomas(P<0.05). the difference of PTEN protein expression was significant between low-grade gliomas and high-grade gliomas (P<0.05).2. the difference of PTEN gene methylation status was not significant between normal brain and low-grade gliomas (P>0.05). the difference of gene methylation status was significant between normal brain and high-grade gliomas(P<0.05). the difference of gene methylation status was not significant between low-grade gliomas and high-grade gliomas (P>0.05). the difference of gene methylation status was significant between normal brain and all gliomas specimens(P <0.05).Conclusions:1. PTEN gene hypermethylation might be involved in the process of malignant glioma.2. PTEN gene methylation may be an important reference for the diagnosis of glioma.
