Objective:To study the in vitro antiproliferative and apoptosis effects of 2,3-indole on renal carcinoma cell line 786-0.We performed surface enhanced laser desorption ionization(SELDI) mass spectrometry using protein chip technology to analysis the molecular mechanism of the antiproliferative and apoptosis effects of 2,3-indole on renal carcinoma cell line 786-0.To find a novel drug targeting of anti-RCC.Methods:Renal carcinoma cell is 786-0 cell lines.1.Inhibitory assay:After being treated with 100~800μmol/L 2,3-indole for 24~72h,the proliferation activities of renal carcinoma cell line 786-0 were studied by MTT and Light microscopy.2.Apoptotic assay: Hoechst33258 Fluorescence microscopy were used to detect apoptosis of renal carcinoma cell line 786-0.3.SELDI assay:We preformed surface enhanced laser desorption ionization(SELDI) mass spectrometry using protein chip technology(WCX2) to compare the changes of protein expression between the medicine group and the control group in vitro.Protein chips were examined in PBSII-C protein chip reader,the protein profiling was collected by proteinchip software3.1 and analyzed by Biomarker Wizard software.Results:MTT assay and Light microscopy showed that the growth of renal carcinoma cell line 786-0 was inhibited by 2,3-indole in a time dependent and dose dependent manners in vitro.Hoechst33258 Fluorescence microscopy showed apoptosis of renal carcinoma cell line 786-0 can be induced by 2,3-indole in vitro.The results of SELDI showed that 4 distinct proteins were found between the medicine group and the control group,with3proteins up-regulated and one protein down-regulated in the medicine group. Distinct proteins were not found in the different medicine group,only with peak intensity different.Conclusions:2,3-indole could significantly inhibit the growth of renal carcinoma cell line 786-0 by blocking cellular proliferation and inducing apoptosis.The mechanism of antiproliferative and apoptosis effects is associated with these proteins different expression.SELDI ProteinChip platform was a simple,sensitive and repeatable method.
