AIM:To study the effect of extractive from Chlamys farreri on the anti-tumor and the synergism on the anti-tumor action of CTX.MATHODS:Inoculated 0.2 mL H22or S180 tumor fluid into the right armpit of each mouse.Animals were divided into seven groups randomly:control group;positive roup(CTX 10 mg.kg-1);high,medium and low dose(1000,500 and 250 mg.kg-1) extractive from Chlamys farreri groups and groups of extractive from Chlamys farreri of high and medium dose used together with CTX.,extractive from Chlamys farreri were given by intraperitoneal injection or intragastric administration for 10 days,Each mouse was given 0.5mL. CTX were given by intraperitoneal injection for 10 days,Each mouse was given 0.2mL.Same volume saline were given to the control groups.Mice were killed after 24 hours of the last dose ten days later and the rate of tumor inhibition were calculated.The acute toxicityof extractive from Chlamys farreri was detected by intravenous injection and intraperitoneal injection.The mutagenic action of extractive from Chlamys farreri were observed through the Ames test,micronucleus test and chromosomal aberration test.Long-term toxicityof extractive from Chlamys farreri was detected by hematological and biochemical indices and organs histopathology RESULTS: The high,medium and small dose PCF had no effect on tumor inhibition of H22.The PCF of high and medium dose used together with CTX produced significance effects on tumor inhibition,the rate of tumor inhibition were 55.9%and 52.9%respectively.Toxicological results indicate that the extractive from Chlamys farreri has no influences on spontaneous movement frequency of mice,the LD50 of intravenous injection is 3187.001mg/kg The maximum tolerable dose of intraperitoneal injection in mice is over 5g/kg,.the Ames test,micronucleus test,chromosomal aberration test of spermatogonial cell are all showed negative results.Furthermore,Long-term intravenous injection also has no influences on body weight,hematological and biochemical indices.The histopathological results of organs show no changes.CONCLUSION:1000 and 500mg.kg -1PCF can enhance the anti-tumor effcts of CTX.And without obvious side effects,.This study provides the foundation in exploration of this potential marine drug resource for further application.
