OBJECTIVE We duplicated the apoptotic model of HaCaT induced by UVA to investigate the impact of UVA on expression of tumout necrosis factor related apoptosis inducing ligand(TRAIL) and the role of TRAIL in UVA-induced apoptosis of HaCaT cells.To study the influence of Polypeptide from Chlamys farreri(PCF) on Fas and TRAIL apoptotic pathway induced by UVA.METHODS Cells were divided into five groups:control group,UVA model group, UVA+5.69 mmol·L-1 PCF group,UVA+2.84 mmol·L-1 PCF group,UVA+1.42 mmol·L-1 PCF group.Using agarose gel electrophoresis,the effect of TRAIL neutralization antibody on UVA-induced apoptosis was investigated.Expression levels of Fas and TRAIL mRNA was assayed by PCR.Western blot analysis was used to determine the protein levels of Fas,TRAIL,FADD and caspase-8 activation.RESULTS TRAIL mRNA and protein levels increased after 8J·cm-2 UVA radiation and the discrepancy is significant compared with control group(P<0.01).TRAIL neutralization antibody had inhibitory effect on UVA-induced apoptosis of HaCaT cells. 1.42-5.69 mmol·L-1 PCF inhibited UVA-induced mRNA and protein expression of Fas and TRAIL in HaCaT cells dose-dependently(P<0.05,P<0.01).PCF attenuated UVA-induced FADD protein expression and caspase-8 activation in dose-dependent manner.CONCLUSION UVA enhances TRAIL expression in HaCaT cells.TRAIL is involved in UVA-induced apoptosis.PCF could attenuate UVA-induced Fas,TRAIL and FADD expression and caspase-8 activation.Its anti-apoptotic effect is related to the effect on Fas/TRAIL-FADD-caspase-8 pathway.
