| Objective:According to the cognition of Chinese traditional medicine to the etiological factor and disease mechanism of stroke sequela,adopt multiple factor to replicate the stroke sequela models of rats which are lack of Qi and with blood silted, and observe effect of BYHWD on expression of Vascular Endothelial Growth Factor in brain tissue of the stroke sequela models of rats in order to offer evidences for establishment of model and the pharmacology effects of BYHWD.Methods:1,Replicated the stroke sequela models of rats with deficiency of qi and blood stasis:The model was established by means of craniectomy in which the left common carotid artery was light with 4-0 silk thread followed by cauterization of the right middle cerebral artery,and the right common carotid artery was occlusioed 30min. Programmed electric shock was administered 24h after the ischemic stroke for 2h daily for 15 consecutive days.2,The effect of BYHWD on pathology change of the stroke sequela models of rats with deficienry of qi and blood stasis:54 wistar rats were randomly divided into normal group,sham surgery group,model group,high-dose BYHWD group, mid-dose BYHWD group and low-dose BYHWD group.The rats in the treatment groups were fed with BYHWD by gastric tube once daily for 15 consecutive days whereas normal and sham surgery group were received the same volume physiological saline solution.The neurological signs of rats were evaluated with the method created by Longa EZ.The pathology morphological changes were determined through TTC and HE staining.3,The effect of BYHWD on the permeability of Blood-brain Barrier of the stroke sequela models of rats with deficiency of qi and blood stasis:96 wistar rats were randomly divided into normal group,sham surgery group,model group, high-dose BYHWD group,mid-dose BYHWD group and low-dose BYHWD group. The rats in the treatment groups were fed with BYHWD by gastric tube once daily for 15 consecutive days whereas normal and sham surgy group were received the same volume physiological saline solution.Half of each group was measured total tissue water content in brain,the other was measured the permeability of Blood-brain Barrier by Evans Blue.4,The effect of BYHWD on the expression of VEGF of the stroke sequela models of rats with deficiency of qi and blood stasis:96 wistar rats were randomly divided into normal group,sham surgery group,model group,high-dose BYHWD group,mid-dose BYHWD group and low-dose BYHWD group.The rats in the treatment groups were fed with BYHWD by gastric tube once daily for 15 consecutive days whereas normal and sham surgy group were received the same volume physiological saline solution.The expression and protein level of of VEGF in brain of every groups were measured by immunohistochemisty and ELISA.Results:1,The rats of model group showed typical symptom of stroke sequela.In addition,the sign of lack of Qi and with blood silted which belongs to the Chinese traditional medicine was seen.30d after the ischemic stroke,there was significant difference on the neurological signs scores among all the groups.The scores of model group was obviously higher than high-dose BYHWD group and mid-dose BYHWD group(P<0.01).The difference between low-dose BYHWD group and model group was not significant(P>0.05).It was proved that BYHWD can alleviate the symptom caused by ischemic stroke.The pathologic slice also showed that BYHWD can significantly ameliorate the pathology injury of the brain.2,There was significant difference on the content of water in brain among all the groups.The content of water in brain of model group was significantly higher than the other groups(P<0.05).The difference on the content of water in brain among BYHWD treatment groups was not significant(P>0.05).There was significant difference on the content of EB in brain among all the groups.In comparison with normal group and sham surgery group,the content of EB in brain of model group was obviously higher(P<0.01);in comparison with mid-dose BYHWD group and high-dose BYHWD group,the content of EB in brain of model group was obviously higher(P<0.05).3,There was significant difference on the protein level of VEGF in brain among all the groups.In comparison with normal group and sham-surgery group,the protein level of VEGF in brain of model group was significantly higher(P<0.01).In comparison with BYHWD treatment groups,the protein level of VEGF in brain of model group was obviously lower(P<0.01).There was significant difference on the number of VEGF immunoreactive cells in brain among all the groups.Compared with normal group and sham-surgery group,the number of VEGF immunoreactive cells of model group was obviously higher(P<0.01).In comparison with BYHWD treatment groups,the number of VEGF immunoreactive cells in brain of model group was significant lower(P<0.01).Conclusion:After ischemic stroke,the typical symptom caused by ischemic stroke that reflected the inadequacy of nervous function,otherwise,the damage of Blood-brain Barrier maked the content of water in brain increase that occurred cerebral edema,all of these can aggravate the injury of cerebral ischemia and give obstacle to the recovery of nervous function.During the experiment,we found that there were a few VEGF positive cells and occurred low level VEGF protein expression,though it was better than normal group and sham-surgery group,if according to results of neurological symptom and neurological behavior performance,we considered it was protective expression of VEGF aider cerebral ischemic stroke that can not promote the recovery of nervous function.But aider the treatment of BYHWD,a series of results of models rats were improved,including:outside neurological symptom,neurological behavior performance,pathology injury of the brain,content of water in brain, permeability of Blood-brain Barrier,protein level of VEGF and number of VEGF positive cells.All of these showed that BYHWD can enhance the VEGF expression in rats,alleviate the cerebral edema and ameliorate the permeability of Blood-brain Barrier,which is one of possible anti-ischemic mechanism and effect of advancing recovery of nervous function of BYHWD. |
