Objective:According to the cognition of Chinese medicine to the etiological factor and disease mechanism of stroke sequela, adopt multiple factor compound metheod to replicate the stroke sequela models of rats which are lack of Qi deficiency and with blood silted. These based on former works, and to observe ulterioly effect of BYHWD on the change of motor function and structure of neurons synapse in brain tissue of the stroke sequela models of rats in order to offer the evidences for establish of model and the pharmacology effects of BYHWD.MethodsrReplicated the stroke sequela models of rats which are lack of qi deficiency and blood stasis. According to syndrone of hemiplegia and the exterior representations of qi deficiency and blood stasis, scores of behavior change, the infarct volume of brains and the pathology changes observed from light and electricity lens as the basal indexes of determinant the models coming into existence. BYHWD was split into ZhongFang sort, Qudilong sort, Dangguibuxue sort, and processed with poaching methods. We selected astragalosidelV ferulic acid and amylose as the quality control standards together. Detected astragalosidelV with the eveaporative light scattering detector, Ferulic acid with UV one of high performance liquid chromatography, and amylose with vitriol-phenol. By testing behaviors scores of rats with balance beam in order to estimate refined motor function of rats. The infarct volume of brains and pathology changes observed from light and electricity lens. The electron and microscope and image alalyzing apparatus were used to measure the change on density, interspace, length of the active area and densematerial of postsynapel of neurons synapse in brain tissue of rats.Results: (1) The elementary index of model(1)Symptom: compared with NG group and APG group, SG group besides symptom of the stroke sequela which adduction of left the upper limp was disability, speeling rat cage, body of rat leaned side , adduction of lower limbs was hardly, walking and sping was gammy , the left eye was smaller than right one , and the spermary was shrink. SG group rats hair intensive dark convoluted, depressed, emotional panic thinness, ingestion decrease, operative scar healing retardation, coincidence the sign of lack of Qi and with blood silted which belong to the traditional Chinese medicine .?The infarct volume of brain count: infarct rate of average iniaret volume of rats is 107.44± 11.04mm3, approximately account for 10% volume of brain. This volume accord with volume of electroagulation median ramus of brains.(2) The scores of motor function: scores of motor function of APG group rats was obviously lower than that of NG group rats[(7±0), P<0.01], The scores of motor function: scores of motor function of SG group rats was obviously lower than that of NGgroup rats[(7±0), P<0.01], The scores of motor function: scores of motor function of SG group rats was obviously higher than that of APG group rats[(4.33±1.67), PO.01].(3) The pathology changes of brain: On light photomicroscope, we saw even distributing of neurone, even shape of karyon in SG group. But, in SG group, we saw a majority of metamorphie putrescence of neurone. On electron photomicroscope, we saw many colloid cell of neurone, no vaculole of cytoplasm, even distributing of brain tissue in NG group. But, in SGgroup, we saw sparse distributing of braing tissue, many vacuole of cytoplasm, partial disconnection endoplast.(4) The synapse changes of neurone: In area of lack of blood and in area of hippcampi, synapse of NGgroupN APGgroup and SGgroup of all have shape trait of stimulant synapse, such as, posstsynase have dense electron compact matter, rounded blastula of synapse. Compared with NG group rat and APG group rats , structure of neurons synapse in brain tissue of SG group rats, the dense of synapse was significantly increased[(8.33±0.91)P<0.05], the interspace of the synapseis, arrower[(10.13±0.56)P<0.05],the length of the postsynaptic thickening was longer(P<0.05) and the dense material of postsynapse was thicker[(127.12±10.38)P<0.05] in area of lack of blood of SG group rats, but there was no dominant changes in area of hippcampi of SG group rats (P>0.05) .(5)The apiece split group function of BYHWD:?Quality control standards: The good linear correlation of astragaloside IV was observed from0.24ug to 6.00ug, that of ferulic acid was observed from 0.08ug to 0.24ug, and that of amylase was observed from 0.102ug to 0.610ug.?The symptom changes of apiece using drug group: After postoperative 35days,the symptom of the rats in the ZhongFang groups , were improved greatly.Their weight was gained,their hair was relative bright, and they acted smartly. The symptoms of the rats in the the Qudilong groups, Dangguibuxue groups were promoted too, but they wer relative depressed and their stool was watery. The rats of the model group were more active than before, but the wounds were cured slowly.?After postoperative 35days, scores of motor function of model group and apiece using drug groups was higher than that of 20days, this account for motor function recuperate of rats, scores of motor function of ZhongFang group rats was obviously higher than that of the Qudilong groups and Dangguibuxue groups rats (P<0.05).(|)On light photomicroscope, we saw many normal shape neurone, few artrophic karyon, even distributing of tissue in ZhongFang group, few artrophic karyon, and few normal shape neurone in Qudilong group, a majority of artrophic karyon of neurone in dangguibuxue group.(§)On electron photomicroscope, we saw intact karyon and cytoplasm of neurone, intact mitochordrion, ribosome and nucleolus, and many colloid cell of neurone in ZhongFang group, swelling and breaking neurone, better karyon of neurone, nucleolus dissolved, few ribosome and mitochondrion ,few colloid cell of neurone in Qudilong group,the half-baked karyon of neurone, erose nucleolus, few mitochondrion.?Compared with the qidilong groups and dangguibuxue groups rats, structure of neurons synapse in brain tissue of zhongFang group rats, the dense of synapse was significantly increased [(21.125±1.125) , PO.05], the interspace of the synapse is narrower[(14.683±0.115), PO.05], , the length of the postsynaptic thickening was longer[(118.616±2.316), P<0.05] and the dense material of postsynapse was thicker[(96.546±4.247) , PO.05] in area of lack of blood of zhongFang group rats.Conclusion: This experiment based on literature shading of the stroke sequela and BYHWD, and combined former studing works, adopt multiple factor compound metheod to replicate the stroke sequela models of rats which are lack of Qi deficiency and with blood silted, and to observe ulterioly effect of BYHWD on the change of motor and structure of neurons synapse in brain tissue of the stroke sequela models of rats in order to offer the evidences for establish of model and the pharmacology effects of BYHWD. As experiment results show that adopt multiple factor compound metheod to replicate the stroke sequela models of rats which are lack of qi deficiency and with blood silted accord with basic require of Chinese medicine syndromes, press close to clinic. In this study we selected a stragaloside IV, ferulic acid and amylase as the quality control standard indexes together. These could essure the stability of the experinments. After stroke, motor function obstacle give priority to refine movement injure of limb far extremity, so, motor function of limb reflected outside syndromes of stroke, obviously reduced motor function of model rats. According to results of thesyndromes, the beam test, and the pathology changes of brain, these shown that the nearone synapse changes of stroke sequela was closely relative to nerve function healing, and this is different from acute stroke, help to determinant stroke sequela models of rats which are lack of qi deficiency and with blood silted. This experiment mainly observed ulterioly effect of BYHWD on the change of motor and structure of neurons synapse in brain tissue of the stroke sequela models of ratso In experiment, we found that BYHWD obviously improved motor function of rats, promoted resume of hemiplegy limb of rats and injuryed nearone synapse of model rats. These changes indicated that BYHWD could improve perfusion of gas and bloods, harmonized Ying and Yang balance, increased bloods providing of organise and apparatus,arosing nerve impulse, and by other ways stimulating pallium, leading to numbel and structure changes of patrescence edge area, thereby, impact on motor function hemiplegy limb. BYHWD could correct metabolize turbulence of nearone which result in by lacking of blood and oxygen, accordingly, alleviated hydrocephalus and the secondary injuring of brain, promoted resume of nerve function, obviously, promoting blood circulation and reliveing blood stasis, especially in Zhong Fang group. The action mechanisms of BYHWD could promote neurone protecting gene of brain neurone livability, keeping function and renew of nearone, these reflected the modern action mechanisms of BYHWD on renewing gas, promoting blood circulation and relievingblood stasis.
