Objective: It has been proved that hemorheology change and the large number of free radicals in vivo accumulation can make fatigue and damage happens, which is one of the great reasons in exercise-induced fatigue. Pentoxifylline (PTX) is an alkaloid extracted from cacao which has high biological activity. Owning to its extensive effect on our body PTX has been widely used in medicine field, but in sports medicine field it is less reported. In this dissertation we studied the ability of anti-fatigue ability of the compound PTX by the measurement and analysis of the hemo-rheological influence and the oxidation resistance ability of PTX hoping to offer some experimental and theoretical data for the research and application of PTX in sports fields.Materials and Methods: (1) Exercise training: running. The objects in this research are 32 healthy male SD rats provided by the Animal Laboratory Center of Nanchang University and they were divided into four groups at random by weight: A, Quiet group(8); B, Exhausting exercise group(8); C, Exhausting exercise + dosing the compound Taurine group(8) and D, Exhausting exercise +dosing the compound PTX group(8). It took 30 days for this experiment. From the second week start to dose the rats medicine of the compound Taurine or PTX in group C and D, to dose the same dose of saline in group A and B, and implement increasing loading running exercise in group B,C and D since the second month. (2) Fatigue Model: exhausting swimming training. After 3 weeks exercise training and medicine, the exhausting swimming experiment was started and the rats were killed instantly after exhausting exercise to take blood and tissue samples for measurement. (3) Histological observation:rat's aortic endothelial cells(AEC)'s cultivation. Four groups were divided: A, the normal group which not any PTX or oxygen free radical damage medicine(ORM) was added to; B,The radical damage group which oxygen free radical damage medicine was added to but no PTX; C, PTX+ORM group1, which 1μg/ml PTX + ORM was added to; D, PTX+ORM group2, which 10μg/ml PTX + ORM was added to and E, PTX+ORM group3, which 100μg/ml PTX + ORM was added to after four generation's cultivation of AEC. And then breed them for 24 and 48 hours observing the cellular form's change and measureing the upper solution.Results: (1) The compound PTX has no side effect on the weight of the experiment rats. Exercise can significantly inhibit the increase of weight. PTX does not affect the growth and health of experiment rats.(2) The compound PTX can significantly improve the exhausting swimming time, reduce blood viscosity, increase the red blood cell's deformability and change hemorheology.(3) The compound PTX can significantly enhance the activity of SOD and lower the MDA content in serum, heart and brain tissue.(4) The compound PTX can clearly change the endothelial cell's morphology damaged by free radical good, enhance the SOD activity and reduce the content of MDA in endothelial cells.Conclusion: The compound PTX has a lot of positive effect in promoting one's sports ability. It can change hemorheology, increase the anti-oxidant capacity of the system, reduce free radical damage to the body, delaying the occurrence of exercise-induced fatigue. Meanwhile, PTX can significantly improve the morphology of endothelial cells damaged by free radical, enhance the anti-oxidant capacity of the system and reduce damages of cells by free radical. In a word PTX can play a protective role for endothelial cells.
