| Background:It is well-known that the balance of organizations is maintained by proliferation and apoptosis of cells. Once this balance has been broken, it will lead to the emergence of a series of diseases such as cancer..In the area of Otolaryngology Head and Neck Surgery, nasopharyngeal carcinoma (NPC) accounted for more than 80% is common in southern China ,while laryngeal cancer is mostly in northern China.Both of the two malignant tumors are vital for patients .Therefore it is enabled the ENT doctors to do more to understanding the laryngeal carcinoma.In recent years,scientists have carried out a series of researches in nasopharyngeal carcinoma that is the aspect of gene and clinical application. And they have got remarkable achievements on the clinical diagnosis and treatment of NPC,while there are few studies on laryngeal cancer which enable the people dysphonia. With the improvement of living standards and economic,in the high demanding of quality of life ,we do some research on spontaneous and radiation-induced apoptosis of larynx.Purpose: Analyze the best dose of radiation of cancer by detecting the apoptosis and relationship of spontaneous and radiation-induced apoptosis of laryngeal cancer .So we could choose the best dose of radiation for treatment.It is good for patients in reducing the cost and improving the quality of life,especially in the protecting of talking.Apoptosis as a kind of biological parameters is used to identify the sensitive to radiation and more importantly characteristics of tumor can be assessed to provide suitable treatment.Research laryngeal cancer in the perspective of apoptosis and and thus guide the clinical dose and the standard laryngeal cancer radiotherapy through studying the Relationship between Spontaneous Apoptosis and Radiation-induced Apoptosis of Laryngeal carcinoma,observing differences of laryngeal cancer before and after radiotherapy,learning the genetic changes and mechanism of spontaneous apoptosis and radiation-induced apoptosis in laryngeal cancer. For the early patients,we should not only cure the disease but also preserve the various physiological functions.To the other patients,it plays a important role of keeping the patient's pronunciation function and ensuring the effectiveness of cancer treatment.Methods:Pick tumor tissue in indirect laryngos or fibrolaryngoscope before radiation, then use 6-MV X-ray linear accelerator for radiotherapy for treatment and observe the lesions of tumors before and after radiotherapy.In the course of radiotherapy, in the radiotherapy of 10 Gy, respectively (GY), 30 Gy and 60 Gy take the tumor in the throat directly from the surface of cells, and make it to 10% neutral buffered formalin-fixed, paraffin-embedded.It is used of terminal deoxynucleotidyl transferase (TdT)-mediated biotinylated deoxyuridine triphosphate (dutp) nick end labeling (TUNEL).Observe and photograph the apoptotic cells (a total of 200-500 cells) under an optical microscope.We can determine the apoptotic cells according to the morphological characteristics(the smaller non-staining cells, integrity of the membrane but foaming, the emergence of apoptotic bodies lately, shrinkage ,rounding,and falling off of adherent cell; concentration, marginalization, lysis of nuclear membrane in staining cells and separating of chromatins into block / apoptotic bodies)Detect rate of apoptosis of laryngeal cancer in the 27 cases,and analyse the date by SPSS software., T test is first for the normal distribution and followed Q test is for the availability of data.Results:The AR of living tissue of laryngeal carcinoma before and during radiotherapy 10 ,30 and 60GYwas 21 %±0.7 % , 60%±1.5%,42%±1.3%,25%±1.9%respectively ,the radiation-induced apoptotic rate was significantly higher than that of spontaneous apoptotic rate( P < 0.05) . In laryngeal carcinoma, spontaneous apoptosis was positively correlated with the radiation-induced apoptosis.Conclusion:Radiation therapy has become one of important treatments of laryngeal cancer, the correct application of radiation technology can preserve the function .not only cure the disease but also improve quality of life of patients.The purpose of this experiment is that to study the changes in radiosensitivity of tumor cells and to analyze the impact of factors in laryngeal carcinoma though the rate of apoptosis of tumor cells before and after radiation ,and find ways to improve the method of radiosensitization so as to provide patients with perfect treatment and reduce radiation injury.We pick tumor tissue in indirect laryngos or fibrolaryngoscope before radiation, then use 6-MV X-ray linear accelerator for radiotherapy for treatment and observe the lesions of tumors before and after radiotherapy.In the course of radiotherapy, in the radiotherapy of 10 Gy, respectively (GY), 30 Gy and 60 Gy take the tumor in the throat directly from the surface of cells and determine the apoptotic cells according to the morphological characteristics and count out the AR.there are differences in apoptosis rate before and after radiotherapy,and the rate of apoptosis in each group after radiation see differences (P <0.05). However apoptosis does not increases with the radiation dose up, on the contrary, the radiotherapy has significantly decreased but still higher than that before radiotherapy apoptosis rate(P <0.05).Detecting of AR of laryngeal carcinoma cells before and during radiotherapy may be helpful to predict the sensitivity of radiotherapy in laryngeal carcinoma patients. |
