Objective:To explore the different expression of IGF-IR and VEGF-C in Esophageal Squamous Cell Carcinoma(ESCC) and the adjacent normal tissue.Methods:1.The expression of IGF-IR and VEGF-C in Esophageal Squamous Cell Carcinoma tissues from 95 ESCC patients and 20 non-cancer people were detected by immunohistochemistry (Envision).Analyze the Correlation between the expression of IGF-IR and VEGF-C and the clinical pathological parameters,analyze the relationship between the expression IGF-IR and the expression of VEGF-C.Then make postoperative follow-up,analyze the relationship between expression of IGF-IR,VEGF-C and survival rate.According to the material nature,we use the the Kaplan-Meier law and the log-rank examination to compulate survival rate and lifetime comparison.2.The expression of IGF-IR,VEGF-C proteins were examined by immunoblotting(Western-blot) in 20 Esophageal Squamous Cell Carcinoma patients tumor tissue and the adjacent normal tissue.The results of quantitive difference was evaluated and analysed by T-test.Result:(1) The positive detection rate of IGF-IR and VEGF-C in ESCC were 81.05%(77/95) and 67.37%(64/95) respectively.And the rate in the adjacent non-cancer tissue was 20.00%(4/20) and 5.00% (1/20) respectively.There was statistical significance between cancer and non-cancer tissue.(P<0.05)(2) The positive detection of IGF-IR was strongly associated with the lymph node metastasis,histological grade and depth of invasion of ESCC(P<0.05).The positive detection of VEGF-C was strongly associated with the lymph node metastasis and depth of invasion of ESCC(P<0.05).However,no significant correlationship was found between the expression of VEGF-C and histological grade(P>0.05).(3) Spearman analysis of rank relativity showed there was there was an obvious positive correlation betwee between IGF-IR and VEGF-C(P<0.05).(4) The follow-up visit in the patients showed that five-year survival rate was greatly lower in the positive expression of the VEGF-C group than that in the negative group(P<0.05).Meanwhile,the positive expression of the IGF-IR group had a lower 5 year survival rate than the negative group obviously.The 5 year survival rate in the IGF-IR and VEGF-C double positive group is lower than the double negative group obviously.The IGF-IR and VEGF-C relative protein levels in esophagus scale cancer tissue were 0.576825±0.028433 and 0.428650±0.053611 respectively.The IGF-IR and VEGF-C relative protein levels in identical patient normal esophagus tissue were 0.427842±0.021425 and 0.306108±0.040964 respectively. The IGF-IR and VEGF-C protein expression levels in esophagus cancer tissues were higher than that of normal esophagus tissues with statistical significance(P<0.05).Conclusions:High expression level of IGF-IR and VEGF-C in the esophagus scale cancer tissues can possibly promote esophagus cancer cell invation and metastasis;and the IGF-IR expression may reflect the esophagus scale cancer cell differentiation degree indirectly;In the esophagus scale cancer tisssues,the IGF-IR and VEGF-C has the obvious rank relevance which indicated that IGF- IR may have the relationship with the growth factoe like VEGF-C signal conduction,and its interaction can promote the formation of tumor;the IGF-IR and VEGF-C expression levels in the esophagus scale cancer tissues have significance in judgement of prognosis;Our research of the impact of IGF- IR and VEGF-C in the formation,development,infiltration and metastasis in the cancer may provide another prospect in clinical dignosis,therapy and prognosis.
